Effect of Nitrogen and Chloro Choline Chloride (ccc) on Microtuberisation in Potato

Abstract/ Executive Summary:

Two experiments were conducted for in vitro microtuberization in potato. The first one comprised of five levels of N (30, 45, 60, 75, 90 mM) and two levels of CCC (0& 500mg/l) and the second experiment comprised of two level of N (45 & 60 m M) and three level of CCC (300, 500, 700 mg/l). The purpose of the experiment was inducing microtuber efficiently in the leading potato cv. Diamante. The cultures were incubated for 75, 90, 105 days for expt. 1 and 90 and 105 days for expt. 2. Initiation of microtuber was significantly affected by N levels. It occurred most early by 5.83 days after culture at 45 mM N with 500mg/l CCC and 60mM N with 0 and 500mg/l CCC in expt. 1 compared to 6.50 to 13.66 days for the rest N levels. In experiment 2 this parameter for 45 and 60mM N was statistically insignificantly N @ 45 and 60 mM produced the highest number (8.83) of micro tubers per flask compared to 6.33 in 30, 75, 90 mM N. the performance of 75 and 90 mM N was the poorest . in expt. 2 the highest of microtuber was 9.50 at 60 mM N and 500mg/l CCC after and 90 days after culture, which was statistically similar to 9.33 at 45 mM N , 300 mg/l CCC at 90 days after culture. The maximum weight of microtuber per flask was 1206.83 mg at 60 mM N, 500 m/l CCC and 90 DAC which was statistically similar to 1142.33 and 1118.83 mg at 45 mM N, 500 mg/l CCC and 90 and 105 DAC respectively. In expt. 2 weight of microtuber flash-1 was 1223.01 mg at 45 nM N, 300 mg/l CCC and 105 DAC. The microtuber weight for 30, 75, 90 nM N, irrespective of CCC and DAC and also at 45 and 60 nM N, with 0 mg/l CCC, ranged from 292.51-698.83 mg. the average highest weight per microtuber was 148.85 mg at 45 nM N, 500 mg/l CCC and 105 DAC. In expt. 2 it was 151.55 mg for 45 nM N, 300 mg/l CCC and 105 DAC closely followed by 60 nM N, 500 mg/l CCC and 105 DAC. In expt. 1 the maximum effective size microtuber was obtained from 60 nM N, 500 mg/l CCC and 105 DAC (75.83%) closely followed by 45 nM N, 500 mg/l CCC and 105 CCC (74.0%), where as in expt.2 it was 74.11% from 45 nM N, 500 mg/l CCC and 105 DAC closely followed the same N level, 500 mg/l CCC and 105 DAC. N @ 45 nM N, 500 mg/l CCC was found the best for producing potato microtuber within 105 days. The protocol developed has the potential for rapid production of potato micro tubers.

Objectives:

T enhance micro tuber production potential of the potato cv. Diamant using different levels of N and CCC.

Methodology:

In vitro ready stock of micro plants of the potato cv. Diamant was used in this study. Two expt. On microtuberisation in relation ti N and CCC were conducted. In expt. 1, five level of N (30, 45, 60, 75, 90 mM) and two levels of CCC (0& 500mg/l) were used and based on results of expt. 1., two levels of of N (45 & 60 m M) and three level of CCC (300, 500, 700 mg/l) were used in expt. 2. Three to five micro plants were developed in vitro through liquid skaken culture as described by Espinosa et al. at the age of 25 days of the micro plants, the liquid culture media were replaced by tuberisation medium with the help of 50 ml bottle top dispenser. The tuberisation media contained basic MS salts supplemented with 10 mg/l BA and 8% sucrose and; N and CCC were used as per treatment. The amount of N was calculated out from NH₄NO₃, which contains 40.20 mM N. the cultures were incubated under dark and at 19±1C. The experiment was conducted following a CRD. The experiment was replicated thrice and three flasks were included in each replication. Micro tubers were harvested at three different dates: 75, 90, 105 days after culture. For expt. 1. and after 90 & 105 days for expt. 2. Aseptically collected micro tubers were washed with DDH₂O and dried naturally under the hood and then put in sterile paradises, sealed by par film and stored in freezer at 4C until further use. The collected data on microtuber production and grading were analyzed and the means were separated using LSD test.

Achievement/Findings:

From the above discussion it may be concluded that micro tubers potato can effectively be produced using 45 or 60 mM N with 300 mg/l CCC. The minimum percentage of effective size micro tuber (>100mg) can be produced with the above levels of N and CCC and 105 days duration of culture.
