Multiple shoot proliferation was obtained by culturing shoot tips and nodal explants of jackfruit plants in vitro . The best response towards shoot multiplication was achieved by adding 1.5 mg/1 benzylaminopurine (BAP) in the medium. Microcuttings taken from proliferated shoots produced roots on half woody plant (WP) medium with various concentrations and combinations of IBA and NAA. The optimum rooting was achieved at 2 mg/1 IBA. About 80% of these plantlets survived when rooted plantlets W .1 :1 v/v transferred to the soil substrate.

For multiplication of year­ round jackfruit genotype using in vitro technique

Young  leaves, shoot  pieces  and nodes were collected from 2 to 3 months old seedlings grown  in a mist house and then washed with running  tap  water  for  about  30 minutes. The explants were then placed  in  autoclavable  jar with 0.1% HgCI2 solution for 10 minutes and  washed  4  times with sterile distilled water under clean bench. The shoot pieces and nodes were cut  into  1.0 cm  long, and  leaves were cut into 1.0 cm  squares Single explant was cultured in each culture vessel of 30 ml size containing 10 ml woody plant medium (WPM). The culture medium was incubated at 25 ± 200C under fluorescent light (2000 lux) following 16 h day (light) : 8 h night (dark) cycle. The culture medium was composed of woody plant medium with full strength salts , 3% sucrose, 0.8% Difco Bacto agar and different concentrations of cytokinins (BAP, Kinetin) singly or in combination with NAA for shoot proliferation. The pH of the media was adjusted to 5.7 ± 0.1 and the media were autoclaved for 25 minutes at 12100C. The control treatment received no supplements of the growth regulators. Seven to ten days after incubation multiple shoots developed at the base of the explants were subcultured at an interval of 3-4 weeks.Half strength WP salts was used for rooting of shoots . All media were supplemented with 2% sucrose and 0.7% Difco Bacto agar and the pH was adjusted to 5.7 ± 0.1. The rooted plantlets were removed from the culture vessel and agar was carefully washed away and transplanted in small plastic pots containing soil and organic manure mixture (1:1v/v). The potted plantlets were gradually hardened in a mist propagating house through periodic exposures to the low humidity of ex vitro conditions and subsequently established  in the soil.

Among  the three different explants,nodes proved to be the best for multiple shoot proliferation when different concentrations of SAP ( 1.0-5.0 mg/1) and Kn (1 0-5.0 mg/1) were used singly and NAA in combinations with  SAP and Kn at 1.0 mg/1. For inducing proliferation and growth of shoots, WP medium containing  1.5 mg/1 SAP was best followed  by 1.0 mg/1 SAP  in both node  and shoot  tip. However. the shoot  multiplication  rate increased with   the  increasing  number   of subculture when 1.5 mg/1 SAP was used in WP medium and at about 10 weeks , 4-5  shoots  per  node culture were produced. For shoot proliferation, kinetm was found less effective  than  benzylaminopurine NAA in combination with SAP and kinetin inhibited shoot proliferation . It  was  also  observed  that  shoot proliferation from  node and  shoot tip explants in WP medium at 5mg/l SAP or Kinetin exhibited no effect. It  was also   found    that   callus formation was found at base with higher  concentration  (5  mg/1)  of SAP. Among the auxins tested , rooting response of microcuttings cultured on IBA-containing medium was superior than others However the best response was observed when 2.0  mg/1  IBA  was used in Y2 strength WP medium. Both the number and length of roots were increased in IBA treatment. At lower concentration (0.5 mg/1) , IBA and NAA failed to induce root initiation It is interesting  to  note that the microcuttings produced roots in vitro in the absence of auxins But the number of roots per micro-cutting was few in auxin­ omitted medium NAA produced roots but all concentrations tended to accelerate callus formation at the micro-cutting base Higher concentration (2-5 mg/1) of NAA. Plant with sufficient roots when transplanted in pots containing a mixture of soil and organic manures (1 1 v/v), about 80% plants survived and established in soil mixture
