Estimation of Genetic Diversity of Okra (abelmoschus Esculentus L. Moench) Using Rapd Markers

Abstract/ Executive Summary:

Genetic diversity and molecular phylogeny of okra were investigated with RAPD markers. One hundred and twenty one genotypes of okra were used for DNA profiling with Randomly Amplified Ploymorphic DNA (RAPD) markers. Out of 39 primers screened, five gave 38 clear and bright fragments. There was 32 Ploymorphic bands (6.40 diversity/primer) out of 38 reproducible products (7.6 diversity/primer) which were amplified from the selected five primers, corresponding to 86.70% diversity of the amplification bands. The molecular phylogenetic dendrogram of okra was constructed using UPGMA and based on Nei and Li’s similarity coefficient from pair wise comparisons of RAPD data among 121 genotypes. There were eight groups, according to their diversity. A correlation of 0.54 between molecular and morphological data was observed. The results demonstrated that the test genotypes grouping pattern was similar to multivariate analysis. The present study showed that the genetic base of okra can be rated as high diversity and indicated the potential of RAPD markers for the identification and management of okra germplasm for breeding purposes.

Methodology:

A total of 121 okra genotypes collected by Department of Horticulture, Bangladesh Agricultural University (BAU), Horticulture Research Centre (HRC), Plant Genetic Resources Centre (PGRC), Bangladesh Agricultural Research Institute, Bangladesh used for the study. The study was conducted during 2005-06 in the biotechnology laboratory, Department of Horticulture, BAU, Mymensingh, Bangladesh. The RAPD bands were scored visually on the basis of their presence (1) or absence (0), separately for each genotypes of okra and each primer. Two independent persons performed band scoring for more accuracy. The scores obtained using all primers in the RAPD analysis were then pooled for constructing a single data matrix. This was used for estimating Ploymorphic loci, Nei’s (1973) gene diversity, population differentiation (GST), gene flow (Nm), genetic distance (D) and constructing a UPGMA (Unweighted Pair Group Method of Arithmetic Means) dendrogram among populations using POPGENE (version 1.31) (Yeh et al., 1999) computer programme. The same programme was also used to perform test of homogeneity in different locus between populations pairs.
