Genotypic Variation in Shoot Regeneration from Different Explants of Chickpea (cicer Arietinum L.)

Abstract/ Executive Summary:

Five varieties of chickpea cv. Nabin, Deshi, ICCL-83105, ICCL-85222 and RBH-228 were tested with two different callus induction media (CIMl) MS+3.0 mg/1 .2, 4-D+3.0 mg/1 BAP and (CIM2) MS+3.0 mg/1 NAA+3.0mg/l BAP for investigation of response of genotypes on induction of callus and shoot regeneration from different explants. Nabin produced highest percentage (94.0) of callus from cotyledonary node and node explants on MS medium containing 3.0 mg/1 2, 4-D and 3.0 mg/1 BAP. Calli of Nabin formed highest percentage (65.0) of shoots with highest (l0.50) number of shoots per callus on MS+3.0 mg/1 BAP+0.5 mg/1 NAA from cotyledonary node explants. The regenerated shoots were rooted on MS and ½ MS media containing 1.0 mg/1 IBA and maximum rooting response was achieved at ½ MS+1.0mg/1 IBA. Regenerated plants were successfully established in soil after acclimatization.

Objectives:

To select a suitable variety for regeneration of chickpea growing in Bangladesh for ultimate use in genetic transformation experiments.

Methodology:

The five different genotypes cv. Nabin, Deshi, ICCL-83105, ICCL-85222 and RBH- 228 of Cicer arietinum L. were used as experimental materials in this investigation. Seeds of 60 Huda et al. Genetic va these varieties were collected from Bangladesh Agricultural Research Institute, Joydevpur, Gazipur, Bangladesh. Seeds were washed thoroughly under running tap water, then treated with 1% savlon marketed by ACI Ltd. and four-five drops of Tween-80 for about 20 minutes. This was followed by successive three washing with distilled water to make the material free from savlon. Surface sterilization was carried out with 0.1 % HgCh for seven minutes followed by gentle shaking. The material was then washed with sterilized distilled water for five minutes giving four five changes and then removes seed coat for easy to germinate by placing them on solid agar MS medium for growth of seedling. For callus induction explants were prepared from in vitro grown seedlings. Hypocotyl, cotyledonary node, cotyledon, node, internode and leaflet explants of different genotypes were used for callus induction. Explants were transferred to 2SxlS0 mrn culture tubes with ISml media containing MS+3.0 mg/I 2, 4-D+3.0 mg/l BAP (CIMl) and MS+3.0 mg/I NAA+3.0 mg/l BAP (CIM2) for callus induction. pH was adjusted to S.7 prior to autoclaving. Cultures were incubated at 2S±I°C with 16h photoperiod. Callus from these primary cultures were transferred to MS medium containing 3.0 mg/l of 6-benzyl aminopurine (BAP) and 3.0 mg/I Kinetin (Kn) in combination with O.S mg/l a-Naphthalene acetic acid (NAA) and O.S mg/l Indole-3-acetic acid (IAA). Data on shoot proliferation efficiency were recorded after 8 weeks of culture. Proliferated shoots were transferred to rooting media (MS and 1/2MS basal media with 1.0mg/1 IBA) for adventitious root formation.

Achievement/Findings:

The results of the present study demonstrate that in vitro plant regeneration could be possible using calli from all these explants except hypocotyl of different genotypes. Shoot regeneration of cultured calli widely varied with different explants and different genotypes. Among the genotypes studied calli, Nabin formed highest percentage (65.0) of shoots with highest (l0.50) number of shoots per callus on MS+3.0mg/1 BAP+0.5 mg/1 NAA from cotyledonary node explants and this was followed by ICCL-83105 which produced 45.0% shoots with four number of shoots per callus in the same hormonal combination and explants.
