Morphological Variation in Isolates of Pyrenophora Teres Causal Fungus of Barley Leaf Blotch

Abstract/ Executive Summary:

Experiments were conducted to determine the variation in 20 isolates of Pyrenophora teres, the causal fungus of leaf blotch of barley. It was observed that mycelial growth, colony characteristics, sporulation, size, shape and septation of conidia varied greatly among the isolates. Mycelial growth varied from 9.26 mm to 24,67 mm. Colonies were effuse, velvety, effuse to velvety, whitish, light brown, brown, gray and black having regular or irregular margin. Number of conidia/cm² of the colony ranged from 3.36 x 10³ to 122. 12 x 10³Length and breadth of conidia varied from 28.12 u m to 75.69 urn and 10.64 um to 15.04 urn, respectively. Number of cells/conidium varied from 2 to 8. Maximum number of conidia was straight and light brown in colour though a few number have been recorded as slightly curved and deep brown in color.

Objectives:

To determine the variation in isolates of P. teres isolated from barley.

Methodology:

Barley leaves having typical leaf blotch symptoms and infected seed samples were collected from five districts of Bangladesh namely Bogra, Dhaka, Dinajpur, Mymensingh and Netrakona. Pyrenophora teres was isolated from the diseased leaf samples following the tissue planting method as used. Pure culture was prepared by single spore isolation technique and maintained on Potato Dextrose Agar (PDA) in test tubes at 4°C. The collected seeds were placed on three-layered moist filter paper in petridish and incubated following ISTA rules. After 7 days of incubation the seeds were observed under stereobinocular microscope and the organism was purified and maintained with the same technique. The collected isolates of P. teres were designated as Pt-I, Pt-2 ....... Pt-20 based on the method used. The growth study of P. teres was carried out using the method. The PDA plates were inoculated with 5 mm mycelial block at the center of the plate maintaining three replications. After 15 days of incubation at 25°C temperature the radial growth was measured. Moreover, color, compactness and shape of the colonies were recorded. In addition number of conidia produced per unit surface area (cm²) of the colony, length, breadth, septation and color of conidia at 15 days after inoculation at 25°C were determined.
