Morphogenic Responses from the Explants of Pigeonpea (cajanus Cajan L. Millsp)

Abstract/ Executive Summary:

Multiple shoot regeneration via callus induction and organogenesis was achieved from the epicotyl and internode explants of in vitro grown seedlings of Cajanus cajan. Maximum number of multiple shoot formation was obtained by culturing the epicotyl and internode explants on MS medium containing 1.0 mg/l BA and 0.5 mg/l NAA. The micro cuttings were rooted to regenerate complete plantlets after transferring them to the rooting media supplementation with auxins namely, IDA, NAA or lAA. Nevertheless, 0.1-0.2 mg/l IDA and NAA was found to be most suitable for root induction. The in vitro regenerated plantlets were successfully established in the soil under natural environment.

Objectives:

To develop tissue culture techniques for callus induction, plant regeneration and shoot proliferation form seedling explants of pigeonpea, as a prerequisite for taking any initiative for the improvement of this important pulse crop using in vitro techniques.

Methodology:

The seeds of Cajanus cajan L. Millsp. were used as experimental materials. For raising aseptic seedlings, seeds were collected from the mature plants and washed thoroughly under running tap water for ten minutes to reduce the level of surface microorganisms. Surface sterilization of the material was done following treatment of seeds with 1 % Savlon for 10 minutes with constant shaking, washing with tap water to make the materials free from Savlon-foam and finally washing carefully 2-3 times with distilled water. The seeds were then taken under running laminar airflow cabinet and transferred to 250 ml sterilized conical flask. After rinsing in 80% ethanol they were immersed in 0.1 % HgCl₂ for 8 minutes. After thorough washing with autoclaved distilled water and after soaking in sterilized filter paper the seeds were finally implanted on the surface of semisolid MS (Murashige and Skoog, 1962) medium for raising aseptic seedlings. The explants were excised from the aseptically grown two-week-old seedlings. The epicotyls or internodes were then cultured on agar-gelled MS medium supplemented by 0.5-2.0 mg/l BA along with 0.1-0.5 mg/l of NAA, 2,4-D, IBA or IAA. The data on intensity of callus growth, percentage of shoot formation, number of total shoots per culture, number of usable shoots per culture and average length of shoots per culture form different treatments were recorded after 10 weeks of culture initiation. Microcuttings were prepared from in vitro grown regenerated shoots and were rooted in ¹/₂ MS and MS media supplemented by 0.1-1.0 mg/l of NAA, IBA or IAA. All media were supplemented with 3% sucrose, gelled with 0.8% agar (Hi- media, India), adjusted to pH 5.7 ± 0.1 and steam sterilized for 20 min. at 121°C under 1.1 kg/cm² pressure. All the cultures were grown at 26 ± 1°C under 14h photoperiod with light intensity of about 2500-lux.

Achievement/Findings:

Results on the in vitro rooting of microcuttings showed that percentage of root induction and number of roots per shoot cutting were obviously influenced by the, concentrations and types. of auxin. Among the IBA or NAA concentrations tested, rooting response of micro cutting in IBA containing medium was better than that in NAA containing medium, while least rooting response was found with all IAA concentration stested. These findings are in agreement with those observed in Adhatoda vasica, chickpea, Ruscus hypophyllum, Pantago ovata and Cephaelis ipecacuanha.
