In Vitro Plant Regeneration from Tobacco Pith

Abstract/ Executive Summary:

Callus induction and plant regeneration from stem explants of five tobacco cultivars and their F₁s were studied to determine the genotypic variation in cultivars and F₁ hybrids. Murashige and Skoog medium supplemented with 0.1 mg/l of kinetin and 10 mg/l of IAA for callus induction and 0.23 mg/l of 6 Benzyl adenine for plant regeneration was used. Hormone free half strength MS medium was used for root induction. Callus induction from pith of Sumatra, White Burley and Marichi was better in light. Virginia Gold and Harison Special showed better callus induction in dark than in light. Callus induction in F j s was better in light than in dark except for Virginia Gold x Sumatra, Marichi x White Burley and Harison Special x White Burley. These hybrids showed better callus induction in dark than in light. Of five cultivars tested, plant regeneration from light mediated calli was highest in one cultivar. All F₁s showed better plant regeneration in light mediated calli. Reasonable plant regeneration was found 35 days after transfer to regeneration medium supplemented with cytokinin and organic compounds where necessary.

Objectives:

To develop a complete protocol and compare plant regeneration from pith of five tobacco varieties and their F₁_s through callus induction.

Methodology:

Seeds of Nicotiana tabacum, variety, Virginia Gold, Marichi, Harison Special, Sumatra and White Burley were collected from the Tobacco Research Station, Burirhat, Rangpur. The present study was conducted in the net house and tissue culture laboratory of Biotechnology division of Bangladesh Rice Research Institute (BRRI), Joydebpur, Gazipur. Ten crosses were done with five parental cultivars and the cross combinations were, Virginia Gold x Sumatra, Marichi x Virginia Gold, Marichi x Harison Special, Marichi x White Burley, Harison Special x Virginia Gold, Harison Special x Marichi, Harison Special x Sumatra, Harison Special x White Burley, White Burley x Virginia Gold and White Burley x Harison Special. The F₁s seeds and their respective parents were sown in the next winter to obtain the F₁s plants as well as the parental plants. The percentage of callus induction was calculated on the basis of the number of explant plated and the total number of callus induced. The percentage of the plant regeneration was calculated based on the number of calli transferred to regeneration medium and the number of calli produced plantlets.
