Evaluation of Some Advanced Mutants of Rice for Resistance to Bacterial Leaf Blight and Sheath Blight

Abstract/ Executive Summary:

Fourteen advanced induced mutants along with five check varieties of rice were evaluated against bacterial leaf blight (Xanthomonas oryzae pv. oryzae) and sheath blight (Rhizoctonia solani) during four consecutive T. aman seasons from 1994 to 1997. For bacterial leaf blight, flag leaves were inoculated with the causal bacterium (103 cell/ml) by clipping method. For sheath blight, the plants were inoculated by placing the inoculum of Rhizoctonia solani inside each hills at flowering stage. All the induced mutants and the check varieties TKM6, Binasail, BR 9 and BR14 were moderately susceptible to bacterial leaf blight. The induced mutant BINA 6-84-4-115, BINA 14-10-10 and the check variety TKM6 were moderately resistant and other induced mutants and check varieties were moderately susceptible to sheath blight except lratom 24 which was susceptible to both the diseases.

Objectives:

To evaluate some advanced mutants of rice in order to identify the sources of resistant materials that can be used as a commercial cultivar or can be used as resistant parent in hybridization programme.

Methodology:

The experiments were conducted at the Bangladesh Institute of Nuclear Agriculture (BINA) farm, Mymensingh following a randomized complete block design with three replications. For sheath blight, ten hills in each plot were inoculated with the fresh culture of Rhizoctonia solani previously grown on the Potato Dextrose Agar. The inoculation was done at flowering stage of plant growth. The inoculum was placed in each hill near water level and wrapped with a piece of absorbent cotton in order to maintain proper moisture for the growth of the pathogen. In case of bacterial leaf blight, forty eight hours old culture of Xanthomonas oryzae pv. oryzae was used to prepare a turbid suspension having a concentration of 108 cell/ml. Four flag leaves of each of ten hills from the middle of each plot were inoculated with the suspension following clipping method. Disease severity were assessed 14 and 21 days after inoculation for bacterial leaf blight and sheath blight, respectively following the Standard Evaluation System for Rice.

Achievement/Findings:

The average mean severity of bacterial leaf blight (BLB) were recorded on these material ranged from 4.3 to 7.0. The least average mean severity was obtained on the induced mutant BINA 4-39-15-13 and the highest on lratom 24. All the induced mutants and check varieties were moderately susceptible to BLB except ltatom 24 which was susceptible. Mean BLB severity obtained for the tested material during four consecutive T. aman seasons from 1993-94 to 1996-97 were more or less consistent except tor the check variety lratom 24 which showed highly susceptible reaction to BLB during 1993-94 and 1994- 95. This variety showed susceptible reaction to BLB in 1995-96 and moderately susceptible reaction in 1996-97. The results of the present investigation revealed that none of the induced mutants and check varieties was resistant to BLB and Sh.B. However, the induced mutant BINA 6-84- 4-115 was identified moderately resistant to Sh.B and moderately susceptible to BLB. This mutant showed more or less consistently tolerant reactions against these two major diseases of rice. This mutant also showed the best performance with respect to yield, panicle length, number of grains/panicle and early maturity among other mutants and check varieties. Based on the seed yield, quality and resistance to diseases, the mutant BINA 6-84-4-115 can be recommended as commercial cultivation for Bangladesh.
