Effect of Strength of Ms Media on in Vitro Rooting of a Miniature Rose

Abstract/ Executive Summary:

The excised shoots of miniature (cv. Dwarf King) rose was cultured In four different strength of MS media with 1 mg/l IBA to examine the rooting efficiency of rose cultivar. Root initiation started 6 to 17 days after Inoculation. The longest duration required for root Initiation was observed on full strength MS medium and shortest was observed when shoots were cultured on 1/2 strength MS medium. Half strength MS media was found to be more effective In respect of rooting In rose and gave 65% acclimatized plants at the end of 40 days In Soil mixture. When full or 3/4 strength MS salt was used in the media, the length and number of roots significantly decreased and gave 40% acclimatized plants.

Objectives:

To determine the optimum requirement of MS salt concentration in the media for better rooting performance of the miniature rose.

Methodology:

The cultured shoots of a miniature rose (Dwarf king) was used in this experiment to find out the optimum MS salt concentration for in vitro performance of this rose cultivar. After 6 weeks of growth in the MS media (Murashige and Skoog, 1962) for shoot proliferation, randomly selected shoots of approximately 2.5 cm in length were transferred to in vitro rooting media. In vitro rooting was induced by inserting the proliferated shoot in four salts based on 1/4, 1/2, 3/4 and full strength of MS media with 20 g/l sucrose and 7 g/l agar supplemented with 1.0 mg/l IBA. The pH of the medium was adjusted to 5.5±0.1 before sterilization at 120°C and 1 atmospheric pressure for 30 minutes. For growth and development of cultures, the temperature was set at 25±1°C with light intensity of 2000 lux from fluorescent tubes and a photoperiod of 16 hours was maintained by timer. After 18 days, in vitro rooted plantlets were transferred to culture pots containing sterilized coarse sand. Immediately after transplantation, the plantlets were irrigated with fine spray of water. The experiment was arranged in completely randomized design (CRD) with 3 replications. Each replication consisted of 20 culture vessels. At weekly intervals, the number of root and the average length of longest root were recorded in a six-week culture period. Days to root formation and percentage of shoot forming root were also recorded. Data were analysed by analysis of variance using LSTAT, a statistical programme. The significant difference between the pairs of means was evaluated by least significant difference (LSD) test.

Achievement/Findings:

Days to root initiation from proliferated shoots of miniature rose were significantly affected by MS salt concentration in the media. The root initiation took place 5-10 days earlier in 1/2 and 1/4 strength MS medium than that of full and % strength. The longest duration (17 days) required for root initiation was observed in full strength and shortest (5.80 days) was observed when shoots were cultured in half strength MS medium which was statistically identical with % strength MS media. The highest rooting percentage was also obtained with the concentrations of 1/4 to 1/2 strength MS salt used in the media.
