Water quality parameters and microbiological aspects of water and bottom sediments in five ponds of different Golda (Macrobrachium rosenbergii, de Man) farms under Fulpur of Mymensingh region were investigated. During the study period (August- September/2004) water temperature ranged from 29 to 33°C, dissolved oxygen from 2.28 to 4.13 mg/L, pH from 6.65 to 7.94, alkalinity from 44 to 70 mg/L and chlorophyll a concentration from 61.88 to 102.34 úg/L. Among the water quality parameters studied, DO and chlorophyll-a levels were found below the acceptable limit. The Aerobic Plate Count (APC) of the water sample was in the range of 1.3 x 104 to 4.29 x106 CFU/g and that of sediment sample was 2.0 x 106 to 2.95 x 107 CFU/g. The total viable bacterial count of pond sediment in the farms indicated the pres nce of high loads of bacterial population. In the qualitative study, some harmful pathogenic bacteria such as E. coli and Salmonella were isolated from pond water and sediment.

The present study reports the various important physico-chemical parameters of pond water quality as well as qualitative and quantitative bacterial standard of water and sediment of Golda (Giant freshwater prawn) culture pond in Fulpur area under Mymensingh region.

Water and soil samples of five freshwater Golda farms under Fulpur upazilla were used for this study. These farms are lsrafil Fishery Ltd.-1 & 2, Helal Fishery Ltd., Samsul Fishery Ltd. and Saidul fish & Golda farm designated as farm I to V, respectively. All these farms are small to medium sized commercial fish farms with round the year production of both Golda and white fish. Important water quality parameters like temperature, pH, alkalinity, dissolved oxygen, chlorophyll-a etc. of various Golda culture ponds were determined. Physical quality parameters of water except temperature were determined in the Water Quality and Pond Dynamics Laboratory, BAU. Water samples (250 ml) were collected carefully in clean plastic bottles and mouth of the bottles were closed with stopper and airtight cap and placed into insulated box. Water samples were collected in the morning, between 9.00 and 10.00 am, and the bottles were filled up with water from various spots and at various depths in each pond. In the laboratory water samples from each bottle was filtered through glass fibre filter paper (Whatman GF/C) and used for analysis. All the analyses were done in triplicate .Bacterial isolation was done by step by step culture of bacteria using a number of selective media. These media include Tetrathionate broth base, Fluid Selenite cystine broth Salmonella-Shigella Agar media, Xylose Lysine deoxycholate agar, and Triple Sugar Iron agar. The inoculated plates and tubes containing different media were incubated at 37°C for 24-48 hours. Different types of colonies which grew on surface of different media were identified by study of morphological and biochemical characteristics.

During the experimental period (August-September/04), water temperature ranged from 29° to 33°C in various Golda culture ponds under investigation. The values were more or less similar in all ponds. This temperature range was suitable for growing of fish and Golda. The temperature recorded in Golda ponds was within the acceptable range for pond fish culture. The main causative agents isolated and identified from pond water and bottom sediments were Streptococcus, Bacillus, Escherichia coli, Salmonella, Staphylococcus and Pseudomonas. Probable sources of transmission might be cross contamination, sewage, fertilization of organic manure and run off from adjacent land. Among these, most significant hazardous bacteria are Escherichia coli, Salmonella, Staphylococcus and Streptococcus.Salmonella was isolated from the sample of Farm I, II and Ill. Regarding the qualitative study of bacteria in farm pond sediments and water Escherichia sp. was commonly found in all farms and Bacillus sp was identified only in farm Ill in addition to Streptococcus.  Presence of coliform bacteria was expected to be very high in the bottom sediments due to presence of huge organic matters on the farm pond bottom because of application of supplemental feeds but their load was moderate. Besides microbiological hazards, there might have some other hazards from pesticides, herbicides, fungicides, antibiotics, detergents, heavy metals etc. that are needed to be studied.
