A study of hematological parameters in the peripheral blood of Thai Pangus Pangastus sutchi was undertaken to identify various blood cells and to carryout some important blood parameters. All of these data were compared with data of fish artificially infected with a pathogenic bacteria Edwardsiella tarda. Different blood cells like erythrocytes, lymphocytes (large and small), thrombocytes, monocytes and neutrophils were identified. Some important haematological parameters were determined such as haematocrit values, haemoglobin concentration, erythrocyte sedimentation rate (ESR), total erythrocyte count (TEC), and total leucocyte count (lLC). Absolute values, viz., mean cell volume (MCV), mean cell haemoglobin concentration (MCHC) and mean cell haemoglobin (MCH) were worked out from the above parameters. Haemoglobin values varied with sex and season. Male showed higher haematocrit value, haemoglobin concentration and erythrocyte count than female. However, ESR and MCV were higher in female than male but other parameters did not show variation. Haemoglobin concentration and haematocrit value showed increasing trend during May to July. Leucocyte count increased during May - June and sharply decreased in July.

a) To know the different types of cells found in the peripheral blood of Thai Pangus (Pangasius sutchi) by characterization and identification using morphological and cytochemical techniques

b)To study different haematological parameters such as total erythrocyte count (TEC), total leucocyte count (TLC), differential leucocyte count (DLC), erythrocyte sedimentation rate (ESR), packed cell volume (PCV) or haemotocrit value, haemoglobin contents (Hb%), mean cell volume, mean cell haemoglobin (MCH) and mean cell haemoglobin concentration (lv!CHC); and

c)To determine the changes on different haematological parameters after artificial infection by Edwardsiella tarda bacteria

 

As an experimental fish, Thai pangus (Pangasius sutchi) (Linnaeus) was selected for this study. In total 66 fish were used as experimental animal. In each month 11 fish were sampled randomly. The sampled fish were collected from Bangladesh Fisheries Research Institute (BFRI) Mymensingh. They were kept in a small pond which was situated behind the wet laboratory of the Faculty of Fisheries and were transported to the laboratory from the pond in bucket containing water. The average length and weight of the experimental fish were 6.5 em and 27.9 g respectively. Average weight of the male fish was 27 g and female fish, 30 g Acclimatization of fish Fish were acclimatized in aquaria containing tap water for 7 days before experiment with aeration and feeding at alternate day with SABINCO fish meal pellet Seventy percent water was changed every day. The aquaria were set in a room (Laboratory - II) based the fish disease laboratory. Blood collection In order to minimize an error in normal blood values, fish were caught gently in a small scoop net avoiding stress, transferred, into a bowl containing the same water where they were acclimatized. After anesthetizing the fish with 5 ppm quinaldine  blood samples were collected from the caudal vein with a heparinized sterile disposable plastic syringe . To avoid contamination with mucus and water the area of insertion of syringe was wiped with alcohol cotton. Collected blood was gently pressed into a sterilized small glass vial.

The  findings of hematological parameters .were within the range found for other fish species. In the healthy P. sutchi haematocrit value or packed cell volume (PCV) ranged from 37-46% which is similar to the results of 36-50% in C. batrachus (Sardar et al. 1999). McCarthy et al. (1973) found haematocrit values of 30- 49% in rainbow trout Salmo gairdneri. Blaxhall and Daisely (1973) also found haematocrit values ranging from 20-43% in S. trutta. In this study haemoglobin concentration ranged from 6-9 g/100 m1 of blood. Sciffinan and Fomun (1959) found a haemoglobin value of 6.5 g/100 m1 of blood in rainbow trout S. garidneri. Sardar et al. (1999) showed 7.6 g/100 of blood which are more or less similar to the present study. l 50 Average value of erythrocyte sedimentation rate (ESR) of P. sutchi was 2.83 nun/h. Sardar et al. (1999) found the ESR value of 3.06 in C. batrachus and Siddiqui and Naseem (1973) showed the value of ESR as 2.5 mmlh in L. rohita, which are similar to the present results.
