Study on Mycelial Growth in Different Mushroom Species and Spawn Production of Oyster Mushroom in Different Substrates

Abstract/ Executive Summary:

The experiment was conducted to investigate the mycelial colony proliferation of different mushroom species (Oyster, Milky and Button) in different media (viz. PDA, YPDA and YMA) and spawn production of Oyster mushroom (Pieurotus sp.) on different substrates (viz. wheat, rice bran and sawdust). In case of media, the best mycelial growth (8.334 em) was recorded in YPDA media at 16 DAI. In case of variety, Oyster mushroom performed best for mycelial growth as compared with other two varieties. In case of combined effect of media and variety, the best growth was found 12.55 em in Oyster x YPDA at 16 DAI. Regarding spawn production, wheat was found the best substrate for Oyster mushroom followed by rice bran and sawdust.

Objectives:

To observe the mycelial growth of Oyster, Milky and Button mushroom on different media and to find out suitable substrates for spawn production of Oyster mushroom.

Methodology:

The experiment was carried out at the Mushroom House and Tissue Culture Laboratory, Horticulture Demonstration and Training Centre (HDTC), Kewatkhali, Mymensingh during January-June, 2005. In this experiment, Oyster, Button and Milky varieties were used. Potato Dextrose Agar (PDA) media, Yeast Potato Dextrose Agar (YPDA) media and Yeast Mannitol Agar (YMA) media were prepared using potatoes, dextrose, yeast extract and mannitol. For spawn production, wheat, rice bran and sawdust were sieved and dried under the sun. Wheat was washed and boiled for 20-30 min. It was dried in the air by spreading on wire mesh to remove surface moisture. Then it was mixed with CaC0₃ (2%) and chalk powder (1%). In case of rice bran and saw dust, these were mixed with wheat (25%) separately. Then 250 g of each substrate was poured into polypropylene bag and plugged with cotton and sterilized. For the preparation of mother spawn a piece of agar medium (30 cm) containing mycelia was placed separately for each substrate base bottles. After 21 days, the whole substrate became white due to fungal mycelia and then it was ready for spawning)he substrate. For spawn production the wheat substrate was cleaned and mixed with gypsum (2%) and chalk powder (1%). In case of rice bran and sawdust, these substrates were stalked in a heap, watered and wheat bran 25% was mixed separately and all these substrate were filled in polypropylene bags @ 250 g and sterilized. Then 5 g previously ready mother spawn was inoculated into each polypropylene bag and spawn was ready for culture. The diameter of mycelial colony of three species of mushroom was measured in centimeter. Spawn production of Oyster mushroom was measured and data were recorded at 3 days interval and spawning rate was expressed in percentage. The data were statistically analyzed following Completely Randomized Design (CRD). The analysis of variance was performed and means were compared by Duncan's Multiple Range Test (DMRT) test at 1% level of probability for interpretation of results.

From the above study, it was found that among the three species, Oyster was the best followed by Milky and Button for cultivation in Bangladesh and YPDA medium was superior to PDA and YMA media and substrate wheat was the best followed by rice bran and sawdust for spawn production of Oyster mushroom.
