An experiment was conducted during the winter season of 1992-93 in the Department of Genetics and Plant Breeding. Bangladesh Agricultural University, Mymensingh, with a view to determining the suitable dose of colchicine for chromosome doubling. The diploid line, BAU-M/ 91 of B. campestris was used to induce tetraploid by colchicine. Four treatment durations viz . 12, 24, 36 and 48 hours with six doses like 0.0, 0.02, 0.05, 0.1, 0.2 and 0.5% of colchicine were used. The experiment was laidout in a split-plot design with three replications allocating the duration treatment of colchicine in the mainplot and the concentration of colchicine in the unit plot. The results showed quite a high degree of influence of colchicine on various characteristics studied. Seedling characters like percentage of survived seedling, cotyledonary leaf width at 7 days after germination (DAG). number of true le aves at 7 DAG, seedling height at 14 DAG, number of true leaves at 14 DAG sho wed negative response to the higher doses of colchicine at different durations. But percentage of apparently 4x seedling showed positive response at 12 and 24 hours and negative response at 36 and 48 hours at 14 DAG . This negative trend might be due to seedling mortality in this duration. After 28 days of germination seedling mortality was 100% at the treatment of 36 and 48 hours of duration with all concentration of colchicine. Plant height at 28 DAG and reproductive characteristics like pollen fertility (%), siliqua set and seeds per siliqua showed negative response whereas days to flowering, flower bud width/length  ratio showed positive response to the higher doses of colchicine. Percentages of apparently 4x plant based on seedling, vegetative and reproductive characteristics showed positive response to both duration and doses of colchicine. The highest percentage of 4x plant (55.6%) were found at 12 hours on 0.2 & 0.5 % and the second highest (50%) was observed at 24 hours on 0.1 & 0.2% of colchicine treatment. So it is being suggested to use with 0. 2 & 0.5 % for 11 12 hours and 0.1 & 0.2% for 24 hours of colchicine to double the chromosome number of mustard &  rapeseed in general, and the yellow sarson genotype BAU-M/91 in particular .

To determine the suitable dose of colchicine for chromosome doubling.

The experiment was conducted at the field laboratory of the Department of Genetics and Plant Breeding BAU, Mymensingh during October- '92 to March '93. The genetic material was BAU-M/91 (Brassica campestris, yellow sarson). The concentrations of colchicine were 0.0%, 0.02%, 0.05%, 0.1 %, 0. 2% and 0.5% and the duration of treatments were 12 hours, 24 hours, 36 hours and 48 hours. The seeds were treated with respective concentration and duration of colchicine. Split-plot randomized block design with three replications was used for the experiment. The duration of colchicine treatment was placed in the main plot and the concentration of colchicine was placed in the sub-plot. The land was well prepared and fertilized with Urea, TSP and MP at the rates of 90, 11 5 and 70 kg/ha, respectively. Cowdung was applied @ 5 t/ha. The unit plot size was 2m x 1m. Half meter space was kept in between the unit plots and one meter between blocks . The row to row and plant to plant distances were 25 cm and 10 crn, respectively. Necessary intercultural operations were done whenever necessary. The data were collected from randomly selected 10 plants in each plot on the basis of percentage of seedling emergence, days required for germination, cotyledonary leaf width at 7 days after germination (DAG), number of true leaves at 7 DAG, seedling height at 1,4 DAG; number of true leaves at 14 DAG, percentage of apparently 4x seedling at 14 DAG, plant height at maturity, days to flowering, flower bud width/length ratio, pollen fertility, percentage of siliqua set, seeds per siliqua and proportion of 4x plants.
