Humoral Immune Response Against Beta and Epsilon Toxoids of Clostridium Perfringens in Small Ruminants

Abstract/ Executive Summary:

Humoral immune response was studied against beta and epsilon toxoids of Clostridium perfringens in small ruminants. Maximum neutralizing antibody titre of log,0 2.10 was recorded on 21st day post inoculation against beta and epsilon toxoids separately. Neutralizing antibody titre of log ,0 1.80 was observed on the said period when beta and epsilon toxoids were inoculated combinely. Significant (p < 0.01 )rise of antibody production was observed from 14th day to 21st day post inoculation of toxoids. The precipitating antibody was detected by Agar gel diffusion test on the 21st day post inoculation in response to inoculation of the toxoids in small ruminants. Better level of humoral immune response of the toxoids was observed in sheep than the goat used in the study.

Objectives:

To prevent different types of Clostridium perfringens infection in animal, their identification and determination of immune responses against the respective toxoid used for the standardization of the vaccine prepared from Clostridium perfringens toxoids.

Methodology:

One millilitre ampoules of freeze dried beta and epsilon toxoids were obtained from International Laboratory for Biological Standards, Ministry of Agriculture, Fisheries and Food, Central Veterinary Laboratory New Haw, Weybridge, Surrey, England. Reconstitution and preparation of toxoid: An ampoule of each of the reference toxoid is reconstituted according to the methods suggested by International Laboratory for Biological Standards, Ministry of Agriculture, Fisheries and Food, Central Veterinary Laboratory, New Haw, Weybridge, Surrey, England. The reference toxoid is diluted in a diluent consisting of 1 part of aluminium hydroxide gel (2%) and 4 parts of physiological saline. Healthy disease free 5-6 months old sheep and goat were used for the production of immune sera against Clostridium perfrighens beta & epsilon toxoid,s. A total of four groups of sheep and four groups of goat consisting of five jn each group were used for the study. An amount of 2.5 ml of toxoid was inoculated subcutaneously in each of the experimental animal. Beta toxoid was Inoculated in one group of sheep & goat. Similarly epsilon toxoid was inoculated in one group of sheep & goat. Both beta and epsilon toxoids were inoculated combinely in one group of sheep & goat. One group of sheep & goat were maintained as uninoculated control. Blood was collected aseptically for sera from jugular vein at 0 day from each of the experimental animal before inoculation of antigen and on 14th day, 21st day after the inoculation of toxoids and the sera were collected separately from each of the ruminant as per standard procedure. Neutralization test was performed to determine the humoral antibody level in sera obtained from beta and epsilon toxoids inoculation in sheep and goat.Agar gel diffusion test was performed on slides in 1% agar gel containing 8.5% NaCL and 0.08% NaN₃. Both beta and epsilon toxoids were used as antigen and immune sera prepared in sheep and goat were used as antibody.
