Effects of Common Vetch (vicia Sativa) on Some Haematologic and Biochemic Aspects of Rats

Abstract/ Executive Summary:

Some haematologic and biochemic effects of common vetch (Vicia sativa) was evaluated in 12 growing Long-Evans rats of either sex, divided into three groups- control (C), vetch supplemented (S) and vetch unsupplemented (U) group, each consisting of 4 rats weighing 100±10 g. Rats of control group fed with commercial poultry pellet (Quality Feeds Ltd., Dhaka), vetch supplemented group fed with ration contained raw vetch as a source of 10% protein and methionine and vetch unsupplemented group fed with raw vetch without methionine for a period of 42 days. Adequate energy, vitamins and minerals were incorporated to fulfil the requirements of growing rats. Results showed that there was an increase in alkaline phosphatase level (p>0.05) which appeared to be almost doubled in rats fed on unsupplemented diet (U) than those of supplemented and control diets. There was also an increase (p>0.05) in plasma cholesterol in rats fed on supplemented Vicia sativa. There was no significant change in the hemoglobin percentage in the rats of experimental groups as compared to the control group. Similarly, the packed cell volume was also insignificant in all three groups.

Objectives:

1. To determine the hematological (e.g. packed cell volume and haemoglobin concentration) and biochemical parameters.

2. To find out the toxic effects of vetch, if any.

Methodology:

The research work was conducted at the Department of Pathology, Faculty of Veterinary Science, Bangladesh Agricultural University, Mymensing. A total of 12 apparently healthy 50 days old Long-Evans rats of both sexes were used in the experiment. The body weights were between 100 and 110 g. The rats were kept in three specially designed separated cages containing 2 in each compartment of cages. In this experiment three types of feed were supplied to the three different groups of rats, such as control feed, vetch unsuplemented feed and vetch supplemented feed. The "Quality" poultry feed (starter-grower) was used as control feed. The vetch-un supplemented feed was prepared by using the following ingredients: Vetch is deficient of methionine. So, .01% methionine was added to the vetch un supplemented feed to make vetch supplemented feed. The rats were randomly divided into 3 (three) groups comprising 4 rats in each of the group. The groups were designated as control (C), vetch unsupplemented (U) and vetch supplemental (S). The animals were weighed carefully and kept group wise in cages. All rats were kept under close clinical observation for a period of 42 days for the appearance of any abnormality. During the period of feeding, the rats were weighed daily and their intake of feeds were recorded regularly. After feeding for 43 days the rats were killed, before killing they were anesthetized with diethyl ether. The blood were collected in the test tube by cutting the jugular vein, with 1 % Na EDT A as anticoagulant. Plasma was collected by centrifuging the blood at 3000 rpm for 30 minutes. The cholesterol was determine (according to CHOD-PAP- method) at the enzymatic hydrolysis and oxidation. The indicator quinoneimine is formed from hydrazine peroxide and 4-aminophenazone in the presence of phenol and peroxides. This was done according to the instruction of "Human Biochemical and Diagnostic (Germany)". The data were analyzed using student's paired 'f' test with Mstat.

Achievement/Findings:

It is, therefore, concluded from this investigation that ad libitum feeding of vetch with or without methionine failed to produce substantial changes in the haematologic and biochemic parameters. This was a preliminary study on feeding raw vetch in rats. However, the detailed studies on toxicity are needed to determine the effects on different animals.
