Carriage of Plasmid DNA by Antibiotic Resistant Bacterial Isolates Recovered from Various Sources of Water

Abstract/ Executive Summary:

Ten bacterial Isolates obtained from different sources of water were tested for antibiotic sensitivity. The river isolates showed resistance to multiple antibiotics. Isolates from municipal tap water supplies and tube-wells were resistant to single or double antibiotics. DNA was purified and agarose gel analysis confirms that multiple antibiotic resistance Isolates contain plasmid DNA.

Objectives:

To study the antibiotic resistance of E. coli, Shigella and Salmonella species isolated from various sources of water of Bangladesh and determine their acquisitions of plasmid DNA.

Methodology:

The pure culture of the bacterial isolates Shigella dysenteriae (R 5 Sh), Shigella sonnei (R 6 Sh), Escherichia coli (R 7 E, R 21 E), Salmonella typhlmurium ( R 26 Sa), Salmonella paratyphi (R 38 Sa), Escherichia coli (Ta 38 E, Ta 42 E) and Salmonella (T 50 Sa), incubated for 18 hours were emulsified in 1.5 ml of nutrient broth and inoculated in to LB (Luria Bertoni) agar plate containing specific antibiotic. The plates were incubated at 37^oc within 15 minutes. After 16 to 24 hours of incubation, the antibiotic plates were examined for the growth of the inoculum as well as complete inhibition of growth. The plasmid DNA of the bacterial isolates were purified and physically characterized following the method described by Watson (1979). Agarose gel elec trophoresis was used to separate plasmid DNA mediated species or DNA fragments in the DNA purified from the Isolates. A polaroid photograph of the gel was taken using UV light source.

Achievement/Findings:

The analysis of purified 0NA confirms that R 5 Sh., R 6 Sh., R 7 E., R 21 E., R 26 Sa., R 38 Sa., isolates although contain plasmid DNA but R 21 E. migration pattern of bands suggested that it might contain more than one plasmids. The plasmid band pattern of R 5 Sh. appeared some what similar to that of plasmid PBE 269 DNA which was used as control (5.5 kb.). R 38 Sa. migrated in a similar pattern to DNA from R 26 Sa. h is likely that the fastest migrating band represents supercoiled plasmid DNA and the slower migrating band represents nicked circular DNA. I n t his study. the antibiotic resistant pattern of the isolates revealed that the isolates from river water were multiple antibiotic resistant and isolates from municipal tap water
