The Pathology of Experimental Production of Enterotoxaemia in Goats and Its Treatment

Abstract/ Executive Summary:

The causal agent responsible for goat enterotoxaemia (Clostridium perfringens) was isolated and identified. For experimental production of the disease in natural host, all the animals were inoculated intraduodenally with freshly grown culture of Clostridium perfringens in thioglycollate medium. Clinical signs developed within 12 hours of post inoculation were similar to those observed in naturally occurring cases.In the treatment trial, combined therapy with Clostridium perfringens antitoxin, procaine penicillin and benzyl penicillin sodium together with normal saline and vitamin B complex was found 100% effective against experimentally induced enterotoxaemia in goat. Combined therapy of Clostridium perfringens antitoxin and oxytetracycline hydrochloride and Clostridium perfringens antitoxin with sulfonamide preparation were found 75% effective. On the other hand, treatment with Clostridium perfringens antitoxin, combined penicillin preparation and oxytetracycline singly could protect goat enterotoxaemia only 25%, 50% and 50% respectively. Thus in the present study, it was observed that antisera in combination with antibiotic gave better recovery rate than the antibiotics alone.

Objectives:

To experimentally production of enterotoxaemia in goats and its treatments.

Methodology:

Samples were collected from goats either suffering from or died of enterotoxaemi both at BLRI goat farm and different parts of Bangladesh. Impression smears prepared from different parts of intestine & direct smears from the intestinal contents of infected goats were stained with Gram's stain and examined following the procedure of Rahman. The collected intestinal contents were cultured on 5% sheep blood agar and nutrient agar media and the colonies with double zone of hemolysis were also cultured in cooked meat broth and brain heart infusion broth and incubated anaerobically at 37°C for 6 days and 3 days respectively to observe the nature of growth in broth media. Biochemical tests were performed with the isolated bacteria according to the procedure described in Bergey's manual.Isolated organisms were used to test the fermentation reaction with basic sugars. Skimmed milk coagulation test also done. A total of 32 conventionally reared healthy unvaccinated Black Bengal goat kids of 6-12 months age were selected for the experiment irrespective of sex. The isolated bacterial culture was inoculated in to the freshly prepared thioglycollate medium contain,ing 0.5% glucose and incubated overnight at 37°C under anaerobic conditions. The goats were prepared for aseptic surgery. The abdominal cavity was approached and the pyloric area of the abomasum and the first portion of the duodenum were exposed. Then 100 ml of freshly grown inoculum in thioglycollate medium was inoculated into the duodenum through a drip over a period of approximately 10 minutes. The abdominal incision was closed by separate muscles and skin sutures with silk. The goats were observed at 3 hours intervals during the first 24 hours post inoculation and subsequently at 6 hours intervals.

Achievement/Findings:

The findings also correlated with the most recent study results against goat enterotoxaemia reported that combined therapy of hyper immune serum and fortified procaine penicillin was the most effective against goat enterotoxaemia. Although slight variation was observed with the present study showed almost similarity with them. These slight variations might be due to variation of drug preparation and sample size of the treatment group.
