Effect of Different Therapeutic Agents on Dichlorvos Induced Poisoning and Haematological Alteration in Guineapig

Abstract/ Executive Summary:

The administration of atropine alone and in combination with nikethamide afforded 100 percent protection against dichlorvos induced toxicity and lethality and completely reversed dichlorvos induced alteration in haematological parameters i.e. TEC, Hb% and TLC in adult male guinea pigs. On the contrary, the administration of nikethamide alone did not afford any protection against lethal dose of dichlorvos and completely failed to reverse the dichlorvos induced changes in haematological parameters in male guinea pigs.

Objectives:

To evaluate the therapeutic efficacy of antimuscarinic drug atropine alone and in combination with CNS (Central Nervous System) stimulant nikethamide and nikethamide alone against lethal dose of dichlorvos in adult male guinea pigs. In addition, the effect of these therapeutic agents on dichlorvos induced changes on haematological parameters was also studied in guinea pigs.

Methodology:

Nine adult male guinea pigs of nondescript breed and weighing between 300-550 gm were purchased from the International Centre for Diarrhoeal Disease Research, Bangladesh (ICDDR, B), Mohakhali, Dhaka. Prior to the commencement of the experiment, all the guinea pigs were acclimatized to the new environmental condition for a period of one week. During the experimental period, the guinea pigs were maintained on a ration consisting of gram, grasses and water ad-lib. All the guinea pigs were randomly divided into 3 groups each consisting of 3 animals and marked as group I, II & III. Based on the results gathered from a preliminary trial, dichlorvos was administered at a single oral lethal dose (80 mg/kg) to all the guinea pigs. The requisite quantity of insecticide was diluted in water and was administered to animals by means of a capillary tube fitted with a syringe. All the three groups of poisoned guinea pigs were treated with various therapeutic agents. Therapeutic agents were injected at 8-10 minutes (min) after administration of dichlorvos. At this time all the intoxicated animals exhibited severe toxic symptoms such as profuse salivation, eacrimation, restlessness, nasal discharge, diarrhoea, frequent urination, coughing, tremors, muscular fasciculations and respiratory distress. Blood samples were collected directly from heart of the guinea pigs in order to determine the reversal of haematological changes prior to and at different time interval after administration of dichlorvos. Total Erythrocyte Count (TEC), Total Leucocyte Count (TLC), Haemoglobin percent (Hb%) and Packed Cell Volume (PCV) were determined as per method. The data were analyzed statistically between normal and treated values by the well known student's test ('t' test).

Achievement/Findings:

The present results suggest that administration of either atropine alone or in combination with nikethamide is capable enough in reversing the dichlorvos induced toxicity, lethality and haematological alterations in guinea pigs.
