Evaluation of the Accuracy of Pregnancy Diagnosis by BARIum Chloride Test in Zebu Cows

Abstract/ Executive Summary:

The accuracy of pregnancy diagnosis of zebu cows done in the Department of Physiology, BAU, Mymensingh using barium chloride test. Five hundred twenty one animals were selected from the BAU Farm, campus and near by village and were grouped into 4 groups according to their gestation period. The groups were, group-I, 30-35 days, group-II, 50-75 days, group-III, 75-100 days and group-IV, 150-200 days of post service. Ten ml of urine sample was collected at around 6, O'clock in the morning and 5 ml of samples were charged with 6 drops of 1% barium chloride. In case of positivity i. e. in pregnancy the urine remained clear and in case of non-pregnancy a white precipitate developed which settled down on standing. The positivity of the test was varied highest in group-I, 82.22% and lowest in group-IV 38.8%. In case of other groups the accuracy was 65.34% and 52.18% in group-II and III respectively. All these results were confirmed by rectal palpation after 7-8 weeks post service. It was observed that pregnancy diagnosis as early as 30-35 days post service was possible by barium chloride test.

Objectives:

To find out a rapid and reliable method for pregnancy diagnosis in our cattle.

Methodology:

Five hundred twenty one pregnant and 99 non pregnant animals from BAU Farm and adjacent village were used in this study. The animals were apparently healthy and their breeding records were taken from the farm registers and owners statement. The animals were grouped into 4 groups according to their gestation length. Group I comprised of 225 cows of day 30 to 35 post service, Group II composed of 101 cows of day 50 to 75 post service, Group III composed of 92 cows of 75 to 100 day post service and Group IV consisted of 103 cows of 150 to 200 days post service. The non pregnant cows were grouped separately. Ten ml of urine was collected in plastic tubes from each cow at around 5.30 to 6.00 a.m. and was labeled and refrigerated at 4°C until tested. No preservative was added. Barium chloride test was performed within one hour of urine collection. Five ml of urine was taken in a glass test tube and 6 drops of 1% barium chloride solution was added drop wise. The reaction (if any) was evident in 2-3 min in the from of white precipitates. There should not be any detectable precipitate if the cow is pregnant, while a clear precipitate should be formed in the urine of a non-pregnant cow. The experimental animals were closely observed for non returned to oestrus and finally confirmed by rectal palpation 7-8 weeks after insemination. Evaluation of the accuracy of the pregnancy diagnosis was done by calculating percentage. The reliability of Barium chloride test was calculated the formula.

Achievement/Findings:

All these results were confirmed by rectal palpation after 7-8 weeks post service. It was observed that pregnancy diagnosis as early as 30-35 days post service was possible by barium chloride test. It is concluded that the barium chloride test is not only important for early pregnancy diagnosis but also easy to test in field condition where facilities of immunoassay are not available. However, detailed research on this test should be administered further.
