Cold Shock Induction in Live Shrimp Penaeus Monodon During Transportation

Abstract/ Executive Summary:

Experiments were conducted to determine suitable temperature to induce anaesthesia in br ackishwater shrimp (Penaeus monodon). Cold shock induction on shrimp was carried out in three ways, namely direct chilling, stepwise chilling and slow chilling. It was observed that shrimp anaesthetised by direct chilling ( short term cold induction at 15°C and transferred under a spray of chilled water f or 24 hours) resulted in 100% sur vival. A model was developed to define anaesthetic stages of shrimp. The most suitable stage of anaesthesia by cold shock induction in shrimp could also be identified.

Objectives:

To study the efficacy of cold shock in bringing about anaesthesia in marine shrimp (Penaeus monodon) and to find out the most suitable stage of anaesthesia for transportation.

Methodology:

Shrimps weighing 18 to 45 g were obtained from a farm (Syarikat Sungai Laut, in Petaling Jaya, Malaysia) and transported to the laboratory at the Faculty of Food Science and Biotechnology, Universiti Pertanian Malaysia, Malaysia and reared in aquarium containing sea water (35 ppt). The aquarium had been provided with filter (horizontal type) for continuous filtration of water while the water was also agitated through aeration. The water temperature in the aquarium varied between 23.5-25.5°C.Apparatus for keeping shrimps in anaesthetic condition under a spray of chilled water: length of 460 mm, breadth 204 m and height 380 mm . Within the chamber, there were three perspex trays 1n layer havif"!g length of 455 mm and breadth of 297 mm. Each of these tray consisted of 160 holes with a hole diameter of 3 mm. A fourth tray was placed at the bottom with the same size but the number of holes were only 96 with a hole diameter of 9 mm. The outer portion of the holding chamber was provided with outlet hole (diameter21 mm) at the bottom and inlet hole (diameter 1 5 mm) at the top. The inlet hole was connected outside with a plastic pipe to a submersible water pump that was immersed in a small tank containing sea water (35 ppt). The inlet hole was further connected over the top o f the holding chamber with a plastic cylinder tube with holes so as to sprinkle water. A frigomix cooler was used to obtain the required temperature of the water sprinkled over the chamber.

Achievement/Findings:

Shrimps placed for long term cold induction (stage 5, sedation 2) and transferred back in the aquarium showed the poorest result amongst the three experiments. Only 80% shrimps were found to .survive out·of which 70% were in inverted position and 30% were in normal position (stage 1). Long term cold induction and shrimps being placed back under a spray of chilled water ( 16.8-17.5°C) entered stage 5 (sedation 2) showed a better result with 100% survival. However, in this system 85.5% of shrimps were in inverted position and only 14.5% were in nor mal position. Shor t term cold induction (stage 4, sedation 1) of shrimps and transferred under a spray of chilled water produced the best result. There were 100% survival and all the shrimps were in normal stage (stage 1) after 24 hours.
