Micropropagation for Vegetaive Seed Production of Sugarbeet

Abstract/ Executive Summary:

This experiment was conducted to find out suitable protocol and sources of explants for micro propagation at BSRI Lab and Field during the cropping season 2011-2012. Sugarbeet (Beta vulgaris) is a cross-pollinated, heterogeneous and highly self-incompatible crop. Sugarbeet have been introduced in Bangladesh which is the source of almost 40% of the world sucrose and it is a temperate crop, but can be cultivated successfully in tropical and sub-tropical regions. But seed production of sugarbeet is not possible yet. That is why micro propagation is the only way to produce plantlets of sugarbeet. Hypocotyle, epicotyle, cotyledon, leaf, leaf stalk and shoot tip can be used as explant for the production of seedlings through micro propagation which was collected from in vitro germinated seedlings under controlled environment. MS medium supplemented with 2,4-D (0.5mgl^-1) containing BAP (0.5mgl^-1) and MS medium fortified with BAP(0.5 mgl^-1) containing NAA (0.5 mgl^-1) separately. Better response was obtained from MS medium fortified with BAP (0.5 mgl^-1) containing NAA (0.5mgl^-1) in case of shoot regenation. Green shoots were placed on MS medium supplemented with NAA (2mgl^-1) from where better performances of root production were exhibited.

Objectives:

1. To identify the suitable sources of explants for micropropagation.

2. To find out suitable media for micropropagation.

3. To develop tissue culture protocols for micropropagation of Sugarbeet.

4. To harden plantlets for transplanting.

Methodology:

1. Hypocotyle, cotyledons, epicotyle, leaf, leaf stalk and dormant shoots were used as explants for micro propagation of Sugarbeet and cultured on MS medium supplemented with different concentration of auxins and cytokinins.

2. The explants were cut into small pieces (about 1cm long) and then were treated with 1% savlon for 5-6 minutes with constant shaking and washed thoroughly with distilled water.

3. Then the explants were taken under laminar airflow cabinet. Surface sterilization were done with a 0.1% mercuric chloride solution containing two-three drops of tween-80 for five minutes under aseptic condition. Surface sterilized explants were washed four times with sterilized distilled water.

4. Then explants were inoculated aseptically for shooting on MS medium with different concentrations and combinations of growth regulators. Shoots were cultured on rooting media for root production.

5. Rooted plantlets were hardened and transplanted in the soil.
