Effect of Salinity Stress on the Ionic Balance and Tolerance of Rice Genotypes

Abstract/ Executive Summary:

Effect of salinity stress on four rice genotypes (Binatoa, BINA-163, Iratom-24 and BINA-3) studied in glasshouse condign. Plants were grown of nutrient solution of different salinity levels (Control, 5,10,15,20 and 25 dSm^-1). Salinity stress increased Na⁺ and Cl^- ions and decreased K⁺ ions in root, stem and leaf but decreased M⁺⁺ ions in root and leaf only. Ca⁺⁺ content of the plants were little affected. Total dry matter (r=0.969) and grain yield (r=0.943) of the rice genotypes were negatively correlated with the concentrations of salts. However, among the genotypes, Binatoa contained lower Na⁺ and Cl^- ions in leaves and showed lower yield under higher salinity stress. BINA-163 contained less Na⁺ and higher Cl^- ions and showed better biomass whereas less yield under salinity stress. Iratom-24 less Na⁺ ions in roots and higher Cl^- in root, leaf and stem whilst considerable biomass and yield up to medium salinity level. BINA-3 showed its susceptibility to salinity.

Objectives:

The objectives were to study the effects of salinity stress on the ionic balance and tolerance of some advanced mutant rice lines/varieties and to identify characters responsible for salt tolerance.

Methodology:

The experiment in plastic pots using nutrient solution with some mutant lines/varieties (BINATOA, BINA -163, Iratom-24 and BINA-3) was carried out in Kharif-1 season, 1992. Binatoa is a local variety widely known to be salt tolerant. BINA-163, Iratom-24 and BINA-3 developed at BINA are known for their potentiality for higher yield and stress tolerance. The salinity treatments were control or 0,5,10,15,20 and 25 dSm^-1. The concentration of the nutrients were 3 mM KN0₃; 2 mM Ca (NO₃) 2,4H₂0; 1 mM MgS0₄, 7H₂0 ; 0.1 mM FEDTA; 0.5 mM(NH₄) 2HP0₄,7.71 µMH₃BO₃; 3.17 µM MnCl₂; 0.55 µM ZnS0₄,7H₂0 ; 0.31 M CuS0₄ ; 0.13µM Co(N0₃) ₂ ; 0.97 µM Na₂Mo0₄ (Munns, 1985). Each pot contained 4.5 litre nutrient solution. The treatments were replicated into 4. The solution was changed once a week and volume of solution was maintained by adding water 2-3 times in a week using weighing method. At the last stage of vegetative growth the plants were harvested. Root, stem and leaf samples were analyzed for Na⁺ , K⁺, Ca⁺⁺, Mg ⁺⁺ and Cl^-. Na⁺ K ⁺, Ca ⁺⁺, Mg ⁺⁺ were analyzed by atomic absorption spectrophotometer and Cl^- was titrated against AgNO₃. At maturity, total dry matter (TDM), yield and 1000- grain weight w ere also recorded. Then the data were analyzed according to two factor experiment in CRD.
