Response of Lentil to Bradyrhizobial Strains in Relation to Nitrate Reduction and Energy

Abstract/ Executive Summary:

A pot experiment was conducted with three promising mutants of lentil (ML-432, N₃M-331 and N₄M-423) and three Bradyrhizobium leguminosarum strains (BINA LT-634, BINA LT 638, BINA LT-640) in sand culture to test the strain specificity and energy status of lentil for NO₃- reduction under nodulation. ML-432 and N₃M-331 responded to all the three strains with spectacular yield enhancement. N₄M-423 responded to strains BINA LT-638, BINA LT-640 and mixed inocula. Strains BINA LT-638 and BINA LT-640 responded unequivocally to all the test mutants. The mutants ML-432 and N₄M-423, NR was lower in both leaf and nodules but sucrose supply enhanced the activity of NR, indicating energy limitation in these two mutants. A single specific strain of inoculums is better than mixed inocula and that energy status of a lentil host for nitrogen fixation and nitrate assimilation depends on the specific lentil genotypes.

Objectives:

Three strains of Bradyrhizobium were tested to find out (i) their specificity with three promising mutants lines of lentil, and (ii) the energy status of lentil for NO₃- reduction under nodulation due to different promising strains of Bradyrhizobium leguminosarum.

Methodology:

The experiment was conducted in pot culture with four replications. Seeds of three lentil mutants viz., ML-432, N₃M-331 and N₄M-423 were sown in pots (25 cm x 20 cm) containing 10 kg sand during the winter season of 1996-97 in a split-plot design. Ten days after sowing, seedlings were thinned to two plants per pot and 1 ml of inoculum broth cultureof Bradyrhizobia viz., BINA LT-634, BINA LT 638, BINA LT-640 and mixed inocula (mixture of the three strains) was applied below the surface at the root zone as per treatments of the seedlings. The plants were regularly supplied with one-fourth strength sterilized Long Ashton nutrient solution without nitrogen in the medium twice a week until maturity. Both leaf and nodule nitrate reductase were determined. Chlorophyll was also estimated. At harvest, yield components and seed yield was recorded.
