Rejuvenation of Degenerating Sugarcane Germplasm Through In-vitro Culture

Abstract/ Executive Summary:

This experiment was conducted to regenerate the degenerating varieties/clones of sugarcane in the tissue culture laboratory of Breeding Division at BSRI, Ishurdi, Pabna during the cropping season 2011-2012. Different aged somatic or meristemetic tissues were cultured in MS (Murashige and Skoog, 1962) medium and modified MS medium supplemented with growth regulators to produce in vitro plantlets.Under this programme, degenerating sugarcane germplasms were rejuvenated using low concentrated 2,4-D (0.25mg/l) and GA₃ separately..For rejuvenation, in vitro plantlets were developed from the degenerating genotypes of CP 50-70, CoK 31, Co 1158 and I 52-88 and planted in the field. After field evaluation (3 years at G_o, G₁and G₂ generation) 4 rejuvenated lines were selected in respect of their better performance and planted in G₃ generation. Besides, three most prominent rejuvenated lines of Co 1158 (0.25D), 82-1089 T₁ and Isd 5-55 T₂ are being maintained at G₅ generation for next year breeding trial.

Methodology:

Degenerating varieties of germ plasm bank of BSRI were included in this programme. The experiment were carried out in the tissue culture laboratory of Breeding Division at BSRI, Ishurdi, Pabna. Different aged somatic or meristemetic tissues as explants were collected from field grown degenerating plants at different times. General aseptic techniques concerning in vitro plant tissue and organ culture were followed. The explant were cultured in MS (Murashige and Skoog, 1962) medium and modified MS medium supplemented with growth regulators to produce in vitro plantlets. Later on, plantlets were potted in glasshouse prior to acclimatization in natural conditions. Finally, newly developed plantlets were transplanted in the field and data will be recorded for comparison with the donor plant.

Achievement/Findings:

After evaluation of in vitro regenerated plants, 4 rejuvenated lines were selected from the degenerating parents and planted in 4m ´ 4m stage at G₃ generation for further evaluation. Besides, three prominent rejuvenated lines of Co 1158 (0.25D), 82-1089 T₁ and Isd 5-55 T₂ are being maintained at G₅ generation for next year breeding trial.
