In Vitro Preservation of Sugarcane Germplasm (2011-12)

Abstract/ Executive Summary:

This experiment was conducted to preserve the genetic materials in in vitro condition in the germplasm laboratory of BSRI, Ishurdi, Pabna during the cropping season 2011-2012. For in vitro short-term preservation the Mannitol (5%) and Sucrose (5%) supplemented MS medium showed effective performance to preserve explants under refrigerator condition upto12 months. The shoot tip explant were preserved up to 365 days of survivability on Mannitol supplemented MS medium under dark condition. In this reporting period the shoot tip explant of six genotypes were cultured for preservation. Among the cultured explants of CP 50-70 and Col 66 were found to survive on preserved medium for 365 days and rest of them were survived up to 270 days. The preserved explants of both the genotypes showed the resumption of growth on shooting medium and after acclimatization the regenerated plantlets were planted in the field at G₀ generation.

Objectives:

I. To preserve the genetic materials in in vitro condition

II. To select the suitable media composition for in vitro preservation

Methodology:

Methodology in this study were carried out in two phases as following:

(1) SOURCE COLLECTION, STERILIZATION AND PREPARATION OF EXPLANT

The explant, which were used as the preserved material, were collected from actively field grown plant. After surface sterilization about 1.0 cm long sprout cutting shoot tips were excised as explant for culture in the medium.

(2) STORAGE CONDITION AND PROCEDURE

In this study for short to medium-term preservation of explant, MS medium containing three protectants at different concentrations and combinations were used. The cultures were maintained both in refrigerator (10-12⁰C) under dark and culture room conditions. After preservation period, explant were regenerated in MS medium supplemented with different hormones.

1) MS +Mannitol + Sucrose

2) MS + Glycerol + Sucrose

3) MS +Mannitol + Glycerol + Sucrose
