Radiation Disinfestation of Oil Seeds and Tobacco Leaves

Abstract/ Executive Summary:

Radiosensitivity of Oryzaephilus surmamensis. Siotroga cerealella and Lasioderma serricorne were studied to disinfest oil seed and tobacco leaves. Eggs were most susccptible. Adults did not emerge when eggs were treated with 0.2 kGy and above. Pupae were most radio resistant. 35.6% adults emerged from the pupae of L. serricorne and 70.5% - 78.4% adults emerged from pupae of 0. surinamensis and S. cerealella when treated with 0.5 kGy. All the three insects were completely sterilized when treated with 0.15 kGy and above. A dose of 0.4 kGy may be recommended tor the complete inhibition of metamorphosis of all these insects.

Objectives:

To investigate radiosensivity of different life stages of insects associated with oilseeds and tobacco leaves during storage, thereby suggesting a recommendation dose for commercial application.

Methodology:

Laboratory culture of S. cereal/a and 0. surinamensis were maintained in wheat and Lasioderma serricorne culture was maintamed on tobacco leaves at 28 ± 2°C and 7 5 ± 5% RH in the laboratory of Food and Radiation Biology . The freshly emerged adults of S. cerealella ( 1- 2 days) and 0. surinamensis (5-7 days) were allowed to lay eggs. on the culture media for 3 days. The eggs (0-3 days) , larvae ( 10- 12 days) and pupae (2-4 days) were irradiated to 0.05, 0.1, 0.15, 0.2, 0.25, 0.3, 0.35, 0.4, 0.45, and 0.5 kGy. The radiation was performed in a 29000 Ci cobalt 60 source, installed by the Bangladesh Atomic Energy Commission at a dose rate of 0.82 Gy /h. Untreated samples were always kept for comparison. Five replications of 100 insects were used for each test. In case of Lasioderma serricorne, newly emerged adults (2- 3 days) were allowed to oviposit on tobacco leaves for 0- 3 days. The eggs were collected along with the leaves and kept in jars ( 24 cx 14 cm x 12 cm) to obtain the various developmental stages of the experimental insects. The developmental stages selected for radiation were, eggs (0-3 days), 4th stage larvae (22- 25 days) and pupae (2- 4 days). Two hundred individuals from each stage were irradiated. Each test was replicated four times. After irradiation. the treated insects were kept under same controlled rearing conditions as the stock cultures. The radiosensitivity of the various stages of insects were determined by the emergence of adults from different treated developmental stages. For sterility test 100 insects from each treatment were observed for 10 weeks to find the progeny production. Observations were made at varying intervals ranging from 1- 7 days, depending on the stage of the insects.

Achievement/Findings:

All the three insects were completely sterilized when treated with 0.15 kGy and above. A dose of 0.4 kGy may be recommended tor the complete inhibition of metamorphosis of all these insects.
