100  semen  samples    of  4 different  breeds  of  bulls   were  evaluated for total count of sperm  per  ejaculation,  motility  percentage   and  methylene blue reduction time.   Simple correlation coefficient between  methylene blue  reduction time and total count and that between  methylene  blue  reduction   time   and  motility percentage were calculated   and  a significant  negative correlation {r=-0.59) was obtained.

To find out relationship between total sperm count, motility    percentage and methylene  blue   reduction  time  of the   semen   sample  to be  inseminated for artificial breeding of cows.

Semen collection was made  once a week  from each of  the 4 bulls  belong­ing   to   four   different  breeds   namely Sindhi, Sahiwal, Jersey   and   Holstein­ Frieslan.  The bulls were in regular stud in the artificial insemination Laboratory of  Bangladesh   Agricultural University, Mymensingh and semen  was  collected by artificial  vagina   method    according to principle adopted.   Immediately after collection of   the   semen  pH,  motility percentage and total sperm count  were determined.  Hydrogen ion  concentration was estimated by  Nttrazine   paper soaked   in  semen  and    observing   the color  change  on the   Nitrazine  paper. The total sperm count was  determined by hemocytometer counting chamber method and motility percentage was worked  out  by   actually   counting  the motile sperm from a small drop of fresh semen on a clean glass slide under microscope at 40X magnification.Methyleneblue reduction test was performed  according to method   described. Methylene blue solution was  prepared first   by  dissolving  50 mg  methylene blue  powder  in 100 cc of 3.6%  sodium citrate  buffer   ( Na8C1 H1 072Ha0 ). Then   egg  yolk   citrate   extender   was prepared  by  mixing egg  yolk,   sodium citrate  and  antibiotics   according to Mukherjee and Baner jee (1980).  After that  0.2 ml   fresh  semen  was  diluted with 0.8 ml of egg yolk   citrate    diluter in a 10 mIvial and   thoroughly  mixed. Only  0.1 ml  methylene blue   solution was then mixed  with  the   former  well in the  vial.    In  order   to  provide    an anaerobic condition to the  medium  the vial was sealed hastily  by liquid  paraffin layer of 1 em  thickness.   The solution   was  than   Incubated in  a  water bath at 4ooc and  the   time  taken   by spermatozoa to bleach the solution was noted.

A total of 100 samples   was   tested  during   the  experimental period  from July 1986 to  January  1987 taking  26 samples from each bull of four   breeds. The  average   volume   per  ejaculation. pH.   motility  percentage,   total  sperm count   and   methylene  blue   reduction time   Sahiwal bull sperm with maximum  motile sperm reduced methylene blue dye in shortest possible time.The apparent   differences  in the above average observations were  each subjected  to analysis  of  variance The  above   analysis    of   variances Indicate that  the differences apparently observed between  and  among   individual dual observation of the bulls   were   due to chance  fluctuation excepting only In the   case   of   motility   percentages where   a   significant    difference    was noted  (P <0.01). The significant  nega­tive   correlation  between  total   count and  reduction  time   (r--0.59)  saw found out  and similarly between sperm motility   and   reduction   time.   These results are in agreement  with Mukherjee and Banerjee (1980) and  also  with Van Demark and his  co-workers (1953).  It is therefore  important that  good  quality semen  with  high  motility  percentage and total  count  reduces methylene blue in shortest  possible   time.
