Histopathology of Irradiated Midgut of Adult Hide Beetle Dermestes Maculatus Degeer (coleoptera: Dermestidae)

Abstract/ Executive Summary:

The non-treated midgut of adult Dermestes maculatus consisted of an epithelial layer, a thin basement membrane, an inner circular muscle layer and outer longitudinal muscle fibers. Most pronounced radiation effect on the midgut consisted of histolysis of the epithelium and destruction of the regenerative cells. The injury to the midgut increased with dosage and with lapse of time from treatment. The extent of radiation damage was found to range from moderate cell disruption after treatment with 100 Gy following complete histolysis at 400 Gy.

Objectives:

In the present paper, the effects of gamma radiation were described in the adult midgut of D. maculatus.

Methodology:

Dermestes maculatus was reared on dried kachki fish (Corica soborna Hamilton) in small rearing jars (5x8x15 cm) and Petridishes (9 cm diam.) at 30 ± 1°C and 70 ± 5% R.H. The adult midgut was used for the study of radiation histopathology of D. maculatus. For each radiation treatment, 5 groups of 10 fresh adults were taken in glass vials. Doses of 100, 200 and 400 Gy were administered at a dose rate of 2670 Gy/hour in a Co⁶⁰ irradiator. The treated and non-treated groups were maintained on their food medium until they were dissected in 8% saline solution on the 2nd and 8th day after irradiation. Immediately after dissection pieces of midgut tissues from treated and non-treated beetles were fixed in alcoholic Bouin's fixative for 22-24 hours, dehydrated in alcohol series and imbedded in paraffin. Achievement/Findings:

The effects of 400 Gy on the epithelial cells of the adult midgut was similar but much more pronounced than those observed at 100 and 200 Gy. In most places, the entire epithelial layer was missing and only the muscle layer remained 1 day after treatment The cytoplasm was found as confluent masses in some areas and the very few nuscle were scattered in the cytoplasmic mass. The regenerative cells were absent. The entire epithelial layer was found to disappear 7 days after treatment . Regenerative cells were completely destroyed and only a muscle layer remained which was broken in many places. Radiation effects on the adult midgut of D. maculatus consisted of marked damage to the epithelium and destruction of the regenerative cells. The extent of damage increased with dosage and with lapse of time from treatment.
