Genetics of Bactertal Leaf Blight Resistance in Rice

Abstract/ Executive Summary:

The genetics of resistance to bacterial leaf bilght (BlB) of three breeding lines, namely, BR161-2B-25, RP633-76-1 AND BR319-1-HR11 was studied against the bacterial isolate BX05. Susceptible varieties used for crossing were Dharial, Marichbati and Panbira. Hybrid populations (F1-F3) were clip-inoculated at booting stage using 2-day-old culture of BX05. Disease evaluation 2 weeks after inoculation indicated that bacterial leaf blight resistance in the test materials is controlled by a single recessive gene.

Objectives:

To study the genetics of resistance in order to develop an appropriate breeding strategy.

Methodology:

Three resistant lines, BR161 -2B-25, BR319-1-HR 11 and RP633-76-1 were used for the genetic study. Susceptible varieties used in the crossing programme were Dharial, Marichbati and Panbira. The resistant materials were of the semidwarf type and the susceptible materials were tall, traditional direct seeded upland aus varieties. Crosses were made in October, 1983 and June, 1984. Parents and F1 's were grown and F1 's were confirmed by comparing with their respective parents in November, 1984. The F2's and F3's were grown during November-June and July-November, 1985, respectively. The experimental field was fertilized at the rate of 120:60:40 kg N:P₂0₅:k₂0 with urea, tripple super phosphate and muriate of potash, respectively. A high dose of N was used to induce disease infestation. Urea was applied in three equal splits, 15 days after transplanting, 35 days after transplanting and at panicle initiation. All other fertilizers were applied at the time of final land preparation. The test population sizes were 10 for each parent and F1. For the F2's the population size varied from 230-474 plants and it was 60 for each F3 family rows. Because rice plants are most vulnerable to BLB at the booting stage, the test materials were inoculated following the clipping method (Kauffman et al., 1975) at the booting stage with 2-day-old cultures of BX05 with approximately 109 cells/ml. BX05 is a virulent bacterial isolate commonly found in the north-western part of Bangladesh. Disease severity was evaluated following the Standard Evaluation System (IRRI, 1980) 2 weeks after inoculation.
