Effect of Different Pathotypes of Bipolaris Sorokiniana on Grain Formation of Wheat Cv. Kanchan Inoculated at Flag Leaf Stage

Abstract/ Executive Summary:

Representatives six different pathotypes of Bipolaris sorokiniana were compared for their effect on leaf blight severity and yield contributing characters by inoculating plants of wheat cv. Kanchan at flag leaf stage under epiphytic condition in pot trials. The experiment was conducted in net house. The fungicides Tilt (0.1%) was used as control to protect the plants from leaf blight disease. The pathotypes did not differed among themselves in respect of Bipolaris leaf blight disease which scored from 82 to 83. In case of yield contributing characters the pathotypes differed among themselves that indicates all available pathotypes of B. sorokiniana in Bangladesh are importance in respect of wheat grain formation.

Objectives:

To compare the different pathotypes on leaf blight severity and its effect on yield contributing characters when plants get infection at flag leaf stage.

Methodology:

The representatives six different pathotypes of Bipolaris sorokiniana were collected from Disease Resistance Lab. of the Department of Plant Pathology, Bangladesh Agricultural University, Mymensigh 2202, Bangladesh. and stored in refrigerator at 5⁰c in slants. The collected soil and compost 4:1 were mixed thoroughly and the earthen pots (20 cm dia.) were filled with the mixture of soil and compost. After preparation of the pot, 10 wheat seeds per pot were sown and finally 5 seedlings were allowed to grow. Plants were watered whenever necessary. Three pots were used for each treatment. The selected pathotypes of Bipolaris sorokiniana were transferred from stock cultures and were grown on PDA in petriplates for 10 days at room temperature. Then the Petriplates having Bipolaris sorokiniana were kept under NUV light for 5 days for formation of maximum conidia. Conidial suspension were prepared with distilled water by collecting the conidia from the agar surface of PDA. The conidial suspension was then filtered through cheese cloth to remove mycelial fragments and lumps of agar. The conidial suspension was then prepared to 10⁴ conidia/ml following the CIMMYT method. The inoculation was done buy spraying conidial suspension at flag leaf stage of wheat plants with spore suspension (10⁴ conidia ml^-1) using a self compressed hand sprayer. Just after inoculation, the pots were covered with previously moistened polyethylene bags for 36 hrs for maintaining moisture. After removing the polyethylene bags the leaf surfaces were exposed but watering of pots were continued for supplying moisture to the seedlings. Control treatment was maintained by spraying the plants with Tilt (0.1%) (Hossain et al., 2001). Bipolaris leaf blight (BpLB) severity was recorded following double digit scale. Yield contributing characters were determined following the method of Hossain and Azad (1994) and grading of seeds were done following the 0-5 rating scale as used.

Achievement/Findings:

The grading zero indicates no disease and 5 grade indicates maximum blank point disease symptom on seeds. The pathotypes varied significantly regarding the formation of grains under grade zero i.e. in case of the healthy seed formation. But the formation of disease seeds under different grades did not differ under different pathotypes of Bipolaris sorokiniana. The findings of the present study has clearly showed the importance of Bipolaris sorokiniana on grain formation of wheat. However, detailed study in this regard need to be carried out in different wheat growing areas of Bangladesh.
