Heritable Nature of Colchicine Induced Variation in (rice Oryza Sativa L.)plants

Abstract/ Executive Summary:

A short period of treatment with 0.2% aqueous solution of colchi cine was given to young seedlings of 10 varieties of rice. In the following generation (CTI) the seeds were grown without further treatment, and only the diploids were kept, and in this CTI generation comparisons were then made between controls (2n) and colchicine-veated diploids (C2n) in some important agronomic characters. The treatment induces changes in the capacity for tiller ing, flowering time. straw and grain yield in most of the rice plants. The changes are heritable in nature and observable in the selfed•seed generation following that in which the treatment was given. In evaluating the novel source of variation in some important agronomic characters an overall significantly higher "improvement.' in all the characters were observed.

Objectives:

To investigate the colchicine-induced novel source of heritable genetic variation in 10 varieties I advanced lines I genotypes of rice plants were studied.

Methodology:

The rice varieties/genotypes/advanced lines were collected from the Breeding division, Bangladesh Rice Research Institute, Joydebpur, Dhaka, Bangladesh and rice project of the department of Genetics & Plant Breeding, Bangladesh Agricultural University, Mymensingh, Bangladesh. In all the cases it can be assumed that the ma te rials were pure breeding homozygous lines. On moist filter paper in July 1991 seeds from ten lines of rice plants were placed in petridishes. The petri dishes were placed in a refrigerator at 4°C for one week to aid germination. When the seedlings had grown 2-3cm some of them were treated by total immersion in 0.2 per cent aqueous colchicine for 2 hrs at room tempera ture, and others were immersed in water as the untreated controls. After proper washing and recovery from treatment the two groups of seedling of each line were planted in compost and grown on in plastic trays in a nethouse for two weeks. The survival rate of the colchicine treated material was of the order of 40 per cent. At seven weeks plants were transferred to 6-inch pots in com post and grown on to flowering in a nethouse. Flowering occurred in October '90-November '90 and all the plants were than bagged for self- pollination. The colchicine treated plants were mix aplaid,with a mixture of 2n and 4n tillers, whereas, the control plants were wholly diploid. The plaidy level of the mixop-laid plants were determined by looking at the root tips of young rice plants.
