Screening of Rhizobium Strains for Indigo Plants

Abstract/ Executive Summary:

A study was conducted in the Department of Soil Science, Bangladesh Agricultural University (BAU), Mymensingh during the period January to May 2005 for isolation, characterization and selection of efficient, strain(s) for cultivation of indigo plants. Rhizobium strains were collected from BAU farm and Haji Mohammad Danesh Science and Technology University Dinajpur. Ten strains of rhizobia designated as IR₁, IR₂, IR₃, IR₄, IR₅, IR₆, IR₇, IR₁₁, IR_l2 and IR₁₃ were isolated and their morphological and biochemical characteristics were studied. The isolates were small rod shaped, motile, whitish in colour, optically opaque, viscous in consistency and produced smooth colonies on YMA medium. A pot culture experiment was conducted using 10 isolates. Seeds of indigo plants were inoculated with Rhizobium strains. Results showed that all the strains recorded significantly higher values in all the parameters, The nodule number, nodule weight, root weight, shoot weight, leaf weight Nitrogen uptake by root and shoot, total biomass and total N uptake by indigo plant ranged from 4 to 37 (IR₂) , 10 to 190 mg (IR₂ ,IR₄), 0.49 to 2.04g ( IR₂), 1.67 to 9.75g (IR₄), 1.35 to 6.03 (IR₄), 8 to 36 mg (lR₄), 19 to 129 mg (IR₄), 3.52 to 17.24g (IR₂) and 99 to 450 mg (IR₄). In all cases the lowest value was recorded in the treatment control. Strains IR₂, IR₄ and IR₇ performed better in respect of all parameters studied.

Objectives:

To reduce or replace the use of costly nitrogenous chemical fertilizer is essential which keeps the beneficial role of rhizobium strains

Methodology:

The experiment was carried out in the Department of Soil Science, Bangladesh Agricultural University, Mymensingh, during January to July 2005. Nodules of indigo plants were collected from the Bangladesh Agricultural University Farm. Medium size, healthy nodules were selected preferably grown on the crown zone of the tap rot of Indigo. Typical rhizobial colonies were observed on the growth medium. Rhizobial colonies were of 1.5-2.0 mm in diameter, round with entire margin, convex or raised, semi translucent to opaque, whitish in color were selected, picked up single colonies carefully and restricted over fresh YMA plates for incubation. The colony morphology such as shape, size, surface, margin, elevation, chromogenesis, consistency, odour etc. and the result were recorded. No or weakly absorption of congo red indicated rhizobia where as strongly absorption indicated contaminants. In case of GPA test poor growth indicated rhizobia where as medium to poor growth indicated contaminants healthy, vigorous, plumy and mature indigo seeds were used in the pot experiment. Before sowing, the seeds were soaked in water for 24 hours and then kept over night for breaking the dormancy. An amount of 9 kg soil was taken in each of 33 pots. The experiment consists of 11 treatments as Control= No inoculation, IR₁ = Indigo Rhizobium 1, IR₂ = Indigo Rhizobium 2, IR₃ = Indigo Rhizobium 3, R₄ = Indigo Rhizobium 4, IR₅ = Indigo Rhizobium 5, IR₆ =Indigo Rhizobium 6, IR₇ = Indigo Rhizobium 7, IR₁₁ =Indigo Rhizobium 11, IR₁₂ =Indigo Rhizobium 12 and IR₁₃ =Indigo Rhizobium 13. The experiment was laid out in a Completely Randomized Design with three replications. After 90 days of sowing, the Indigo plants were harvested including intact root system and necessary data were recorded. The plant samples were analyzed for N, P and K following standard methods. Nutrient uptake was measured from the nutrient contents and yield data.
