Ribulose-1, 5-Bisphosphate Carboxylaee/O.xyginase ( RUBISCO ) is the enzyme responsible for photosynthetic CO !•fixation by Ribulose-1,5•Bisphos­ phate (RUBP) in both C a and Cplants. In rice leaves large subunit of RUBISCO was found to be of about 60-kDa molecular size, when total leaf proteins were fra­ ctionated by Sodium-Dodecyi-Sulphate•Polyacrylamide-Gei-Eiectrophoresis (50S­ PAGE), under reducing condition. The enzyme is important becauae its conent and activity determines the photosynthetic efficiency of specially the Ca plants. Rice is Ca in nature. We treated rice seedlings at one months age with various concentrations of Cycocel (CCC); Indole Butyric Acid ( IBA) and Gibberellic Acid (GAa) and measured the amount of.of RUBISCO Content by SDS-PAGE. It was found that all concentration of CCC increased RUBISCO content and the highest content was at 500 ppm. But GA 3 had an opposite effect. Treatment with GAa decreased RUBISCO content at all concentrations. However, application of IBA did not show any significant variation in the content of RUBISCO as com­ pared to control.

To study the effectiveness of various plant growth regulators

In the grill house of the Department of CropLBotany,Bangladesh Agricultural University, Mymenslngh this experiment has been conducted to study the eff ­ ectiveness of various plant growth regul­ators during May to November 1991. ' Seeds of Oryza fatua were cllected (rom local sources in deep water rice grow­ ing area of Rajshahi. Seeds of Binas­ haif and Nazirshail wars obtained from Bangladesh rnstltute of Nuclear Agrlcu­ lture(BINA). Seedlings were raised from surfact sterilized seejs • and ware grown by pot culture. Design of experiment is completely randomized design. <.rops grown by pot culture were sprayed with various concentrations of chlorocholinchloride (CCC). Indo!e Butyric Acid ( IBA ) and Gibberellic Acid (GA 3) at the age of 30 days of the seedlings. After 15days of the tra.atmant [eaf samples were collected from treated as well as control plants. Total leaf prot,.eins were extracted     with     protein extraction buffer(8   M urea,   20% glycerol. 4% SDS, 5%2-   ME  and  125  mM  Tris-HCI, PH 6.8) and ware fractionated  by SDS-PAGE. RUBISCO was identified by   running pure enzyme in the same gel.Sodium-Dodecyi-Sulphate Polyacry­lamide-Gel- Electrophoresis-( SDS­ PAGE): The basic  procedure  was.that of  Laemmli  ( 1970  ).  Polyacrylamide mini slab gels (10.6 x 6 x 0.1 em) were composed   of  2.75%  stacking  gel (1.5 em)  and  15%  separating gel (4.5 em). Samples  were      dissolved        in   laem­ mli'a  sample  buffer   at  95°C  for 3 min and  were   separated with a buffer sys­ tem of 25   mM  tris  containing 192  mM glycine  and 0.1%SDS at room temperat ure. Proteina were run at a constant vol tage of 50 V for 2 hours.
