Induced Chromosomal Aberrations in Diploid and Tetraploid Lolium Perenne L.

Abstract/ Executive Summary:

The effect of different physical and chemical mutagens on the induction of mitotic aberrations indiploid and tetraploid Lolium perenne has been studied. In case of both diploid and tetraploid perennial ryegrasses all the mutagens were found to be effective in inducing mitotic chromosomal aberrations. Mitotic abnormalities were more in the tetraploids as compared to that of the diploids. Chromosomal aberrations were more frequent due to the physical agents than to the chemical agents, which leads to a conclusion that although all the types of mutagens are effective but physical agents are more eff ective in comparison to the chemical agents in the induction of chromosomal aberrations in both diploid and tetraploid Lolium perenne.

Objectives:

To study the effect of different mutagens such as gamma rays, ethyl methylsulphonate (EMS), diethyl sulphate (dES) and ethylene imine (El) on diploid and tetraploid perennial ryegrass was initiated. The present paper deals with the mitotic chromosomal aberrations induced by different mutagens in diploid and tetraploid forms of Lolium perenne L.

Methodology:

Seeds of diploid and tetraploid Lolium perenne of equal size and uniform moisture content were irradiated with 5, 15, 25 and 35 KR of Cobalt-60 gamma rays. They were also subjected to treatment separately with three chemical mutagens, EMS (0.05%, 0.10% and 0.15 %), dES (0.02%, 0.06% and 0.10 %) and El (0.0025%, 0.005 % and 0.0075%) for 24 hours. The seeds were· arranged in petridishes for cytological studies after treatment with the mutagens. Root tips were fixed in 1:3 acetic alcohol and were squashed in hematoxylene using ferric alum as a mordant. The mitctic metaphases and anaphases were scored to determine the percentage of chrorr:osomal aberrations. Different abnormalities such as, fragments, laggards, precocious movement, non- orientation of chromosomes and bridges were scored in the root tip cells of the treated material.
