Nitrate Assimilation in Peanut in Relation to Rhizobial Strains and Carbohydrate Supply

Abstract/ Executive Summary:

The experiment was conducted under sand culture condition with four strains of peanut Rhizobia viz., BINA-GN-205, BINA GN-1000, BINA GN-1317 and BINA GN169 to find out the efficiency for seed productivity and energy relation between nitrate reduction and dinitrogen fixation, revealed that BINA GN-1000 and BINA GN-1371 strains significantly increased pod and seed yield. Data on nitrate reductase activity in leaf and nodule reflected energy limitation at peak-flowering stage and energy saturation at pod filling stage. The energy relation has been discussed in the light of nitrate reduction and nodulation.

Objectives:

The experiment was conducted with some strains of Rhizobium of peanut to find out nitrate reductase activity in leaves and nodules in relation to energy supply and select efficient rhizobial strain (s) for peanut productivity.

Methodology:

The experiment was carried out in pot culture condition in sands with one genotypes of peanut viz., ICGV-91096. Pots contained 10.0 kg sands. The experimental pots were arranged in a completely randomized design with four replicates. After one week of germination of sterile seeds, 1 ml of broth culture (10⁸ bacteria/ml) of Bradyrhizobial inocula viz., BINA-GN-205, BINA GN-1000, BINA GN-1317 and BINA GN169 were provided below the surface at the root zone as per treatments of seedlings. Long Aston complete nutrient solution (-N) in ½ strength was provided in each pot in a week after germination of seeds. Plants were provided with adequate supply of distilled water when needed. Two plants were allowed to grow in each pot. At 80^th and 110^th day, 5mM nitrate (KNO₃) in addition to nutrient solution was provided to the pots and nitrate reductase activity in leaf and nodules were determined after 48 hours following the method of Stewart and Orebamjo (1979) in presence or absence of 20 mM sucrose in the assay. Chlorophyll and Leg-haemoglubin was also determined. The collected data were statistically analyzed following Duncan’s Multiple Range Test using the statistical computer package programme, MSTAT.
