Cytological Changes in Wheat by Chemical Mutagens

Abstract/ Executive Summary:

Effectiveness of three chemical mutagens viz. Hydroxyla, mmonium chloride, Ethylene glycol and Triethanomine in producing chromosomal aberrations in mitotic and meiotic celfs of wheat was studied. A II the mutagens produced significantly higher percentage of aberrant cells than the control. Ethylene glycol and Triethanolamine induced maximum .chrouiosomal aberrations in mitot.c and [meiotic cells respectively.

Objectives:

To investigate the cytological changes in wheat due to use of three chemical mustaches were studied.

Methodology:

A hexaploid wheat (Triticum aestivum L.) variety 'Sonalika' was used as plant material. The three chemical mutagens were Hydroxyla- mmonium chloride (HAC), Ethylene glycol (EG) and Triethanolamine (TEA). The chemical formulae and makers of these mutagens are HONHa Cl ( Evmerck ), CH!(OH).CH!OH (BDH) and C₆HuNOa (E.merck) for HAC, EG and TEA, respectively. Methods: Dry. dormant, healthy seeds were presoaked in 0.2% aqueous solution of each of the three mutagens for 12 hours and then thoroughly washed in tap water to remove traces of chemicals. The control was presoa- ked in distilled water (DW) for the same period of time. These were then germinated in peptidases for mitotic studies. Young healthy root tips of suit a ble sizes were co llected, fixed in aceto-alcoho I ( 1:3 ) and transferred to 70% alcohol after 48 hours. Root tips were stained in haematoxylin following some modifications of Haque et al . (1976), and temporary slides were prepared and examined under micros- cope. For meiotic studies, treated seeds were sown in the field treatmentwise,Pollen mother cells (PMCs) were- collected fro n th i plant'> raised from these treated seeds a nd fixed in Carney's. solution ( 6 ethanal : 3 chloroform: 1 acetic acid) for 48 hours. These were then transferred to 70% alcohol and: kept in a refrigerator until used in the laboratory. PMCs were stained in 2% acetocarmine following standard cyto- logical technique and temporary slides. were prepared and observed under' microscope. Photomicrographs of both mitotic and meiotic cells with different abnormalities were taken.
