Determination of Suitable Hcg-doses for Breeding of Clarias Batrachus in Different Months of a Breeding Season

Abstract/ Executive Summary:

Induced breeding experiments were conducted with 61 pairs of Clarias batrachus in order to determine HCG-doses suitable tor breeding the fish during the months of June, July and August of the same breeding season. Three treatments consisting of single doses of 2 .0, 3.0 and 4.0 IU/g of body weight were tried with a sufficient number of replicates during each of the three months. Criteria studied to determine suitability of doses were rate of ovulation success. Fertilization, hatching and number of spawn obtained. A noticeable variation in breeding response was observed over the period of study. In July and August, a dose of 3.0 IU was found to exhibit the best performances followed by 4.0 and 2.0 IU. However, a dose of 4.0 IU showed the best result in June.

Objectives:

To determine suitable doses for successful breeding of C. batrachus in different months of a breeding season using a single dose.

Methodology:

Healthy broods of C. batrachus were stocked in a pond during the month of March at the Freshwater Station, Fishries Research Institute, Mymensingh. The fishes were feed with supplemental feed composed of fish meal (40%), rice bran (20%) , mustard oil cake ( 20%), wheat bran (15%) and molasses (5%) at the rate of 4 % of the total biomass daily. Sixty pairs of fully mature and healthy brood fish were randomly selected for the present study on the basis of external morphological characters. Three single doses (2.0, 3 .0 and 4.0 IU/g of body weight of fish) of active crude HCG (Sumaach) for both male and female fish were tried over a period of three consecutive months: June, July and August. The doses were replicated with 6-8 sets (each consisting of a male and a female) ranging in weight from 120- 162 g in each of the three months. The efficiency of doses was observed .on the basis of ovulation success and subsequent fertilization, hatching rate and number of spawn obtained. For spawning purposes eggs were collected by manual stripping of female and testes were disected from males 20-24 hours after injection. The eggs were fertilized with macerated testes. Fertilized eggs were stirred with the help of feather and washed by normal saline (0.7%) water before being put in the hatching troughs which receive continuous water flow. The number of eggs put in a hatching though as well as the number of fertilized and hatched out eggs were recorded. Total rainfall during the experimental period and water temperature in the incubation troughs were also recorded.
