Tungro Virus Transmission Ability of the Natural Population of Nephotettix Virescens Under Bangladesh Situation

Abstract/ Executive Summary:

Ten days old seedlings of six rice varieties were exposed to viruliferous insects for 1, 2, 4 and 6 hours. Six subsequent inoculation feedings were made on six different groups of seedlings and transplanted on potted soil. The percent infection in different feedings under each inoculation period was recorded to determine the transmission ability of natural population of Nephotettix virescens. Inoculation feeding period had sound effect on transmission ability of the vector. All the insects were efficient transmitter when allowed them to feed for 6 hours while 93 percent insect was able to transmit the viruses whe n inoculation feeding continued for 4 hours. On the other hand, 74 and 64 percent insects were able to transmit the viruses when inoculation feeding period was 1 and 2 hours, respectively . The vectors were able to transmit the viruses up to 5th subsequent feeding with gradual decrease in transmission rate. Thus , it is concluded that once the insects become viruliferous with tungro viruses can infect several plants without further acquisition of the viruses from the diseased plants.

Objectives:

To determine the transmission ability of natural population of Nephotettix virescens in subsequent feedings as influenced by the time of inoculation feeding.

Methodology:

About 700 seeds from each of IR 24, IR42, IR46, IR 50, IR60 and TN1 were soaked in water. After 48 hours. excess water was drained out and the seeds were incubated for 24 hours. The sprouted seeds of each cultivar were sown in wooden boxes separately. Vector insects, Nephotettix virescens, were collected from the tungro infected rice field. The adults were allowed to feed on tungro source plants for three days to acquire the viruses. The source plants were maintained in the greenhouse. The experiments were conducted in the greenhouse of the Department of Plant Pathology of Bangladesh Rice Research Institute, Gazipur, Bangladesh . Ten days old seedlings were uprooted and cleaned in tap water and placed in test tubes containing small amount of water at the bottom to prevent desiccation. The viruliferous vectors were confined in the test tubes (one insect per seedling). Four inoculation feeding periods viz. 1, 2, 4 and 6 hour(s) was (were) maintained. The viruliferous insects of each group were allowed to feed subsequently on six groups of seedlings and maintained separately. One hundred seedlings of each variety were used for each subsequent feeding. The inoculated seedlings were transplanted on the potted soil (sandy soil:cowdung = 1:1) at the rate of 10 seedlings per pot. A set of healthy plants was also maintained as control. Number of seedlings infected were recorded based on visible symptoms at 20 days after inoculation and percent infections were calculated.
