A Comparative Study on the Type and Level of Inoculum for the Development of Sheath Blight Disease of Rice

Abstract/ Executive Summary:

Two types of inoculum such as sclerotia and mycelial block of sheath blight pathogen of rice were used in different number and amount respectively to inoculate 45-day-old TNI plants to evaluate their comparative performance in disease development. Percent infection as well as disease severity were significantly high when mycelial block was used as inoculum. Moreover, inoculation with mycelial block was easy and less time consuming and inoculation with sclerotia was trouble some and more time consuming. Therefore, in a routine screening programme inoculation with mycelial block is suggested to use as inoculum .

Objectives:

1. To evaluate the effectiveness of sclerotial and mycelial inoculation at different inoculum level in order to employ sequential screening programme.

Methodology:

Taichung Native 1 (TN 1), a rice cultivar susceptible to sheath blight pathogen was used in this Study. Seeds were soaked in tap water for 24 hours. Excess water was drained out and the seeds were incubated for another 40 hours. Sprouted seeds were sown on potted soil. The potted soil was prepared by mixing clay loam soil and well decomposed cowdung ( 1:1). All stubles were removed and the soil was prepared into a good tilt. The pots (Sixteen centimeter diametre) were filled with potted soil. The soils were submerged under water for 24 hours and puddled. Fifteen days old seedlings were transplanted on the potted soil at the rate of two hills per pot and 2-3 seedlings per hill. Water to a depth of two to three cm were maintained. The isolates of the pathogen were collected from the field and were inoculated in PDA medium. The inoculum was transferred to PDA plates for pure culture of the pathogen. Rice straw were cut into small pieces and washed in water. The materials were then taken in conical flask and sterilized in an autoclave at 15 PSI. Five mycelial blocks (one cm dia) of the pure culture was added to the conical flasks and mixed with the straw. The flasks were then placed in the incubation chamber for large scale production of sclerotia of the pathogen. The test plants were inoculated at 60 days af ter transplanting. The pots were arranged into 2 1 groups to inoculate with seven treatments having three replications. Each replication consisted of 20 plants. The treatments were one sclerotium, two sclerotia, three sclerotia, one centimetre diameter mycelial block, two centimetre diameter mycelial block and three centemetre diameter mycelial block . The sclerotia were placed on the uppermost leaf sheath of a middle tiller of each plant. The sclerotia we.e kept moist by placing a water soaked cotton around the tiller and tied loosely. The mycelial blocks were placed in the middle of each hill and were placed in contact with the leaf sheath as mentioned above. The cotton plugs were kept moist upto seven days, and then removed. Total number of tillers and sheath infected was recorded at 15 days af ter inoculation and continued upto 65 days after inoculation at 10 days interval. Percent infection of the tillers and sheath was calculated. Diseased sheath area was also determined by eye-estimation. Individual inoculated plants were scored for severity of the disease.
