Germination and Growth Responses of Maize Under Different Salinity Levels

Abstract/ Executive Summary:

A Laboratory experiment was carried out in petridishes to which QPM cv. Nutricta maize seeds were laid on tissue paper beds to evaluate the effect of different levels of salinity 0 (controI), 0.05, 0.1, 0.2, 0.4, 0.8% NaCl on the germination percentage, germination index, water uptake (%). An identical experiment was also conducted in plastic pots at field level in the kharif season of 1994 where in addition to the above parameters, the length of root and shoot together with their dry weights were measured. Seed germination and germination declined significantly with increasing level of salinity while it retained almost identical to that of control until the concentration was 0.1% NaCl. Water uptake (%) decreased with increasing salinity levels. Root and shoot lengths and their dry weights were markedly affected by NaCl concentrations at 0.2% and more.

Objectives:

To find the adaptivity level under salt stress of QPM cv. Nutricta Maize under field and laboratory condition to see the effects germination and growth behaviour.

Methodology:

Laboratory Experiment : Seeds were laid on tissue paper beds in 12 cm diameter petridish, arranged in completely randomized design, with six treatments replicated five times. Five levels of salinity i.e. 0.05, 0.1, 0.2, 0.4 and 0.8% NaCl solutions, obtained by dissolving NaCl in distilled water, and distilled water as Control were used. In each petridish, 10 healthy seeds of OPM cv. Nutricta were placed on the tissue paper bed and the graded solutions of different salinity were added justt enough to immerse the seeds, partially taking care not to hinder aeration. The seeds were allowed to germinate at room temperature. Germinating seeds were counted at 2 days interval and continued up to 10 days af ter placement on tissue paper. Percentage of germination (GP) was calculated. Germination index (GI) was determined by using an equation. To measure water uptake ( %) , 10 days old seedlings were taken from each petridish and their fresh weight was taken instantly . The seedlings were dried in an oven at 80°C for a period of 24 hours and then water uptake (%) was calculated from fresh and dry weight. Pot Experiment: A pot experiment was conducted growing plants in plastic pots containing 20 kg sandy loam soil in each. Calculated amounts of NaCl were mixed with the soils to raise soil salinity levels to 0.05, 0.1 , 0.2, 0.4 or 0.8 % NaCl. Nitrogen, phosphorus and potassium in the form of urea. TSP and MP, respectively were added to the soil at the rate of 190, and 120 . 120 kg/ha, respectively. Ten healthy seeds of QPM cv. Nutricta were sown in each pot. Plants were thinned and 6 plants were maintained in each pot. Root and shoot lengths and their corresponding oven dry weights (at 80⁰ C tor 24 hours) were determined from plant samples collected at 10 days interval starting from 15 days after sowing DAS and continued up to 55 DAS.
