Electrophoretic Analysis of Soluble Leaf Protein in Reproductive Stage of Rice Plant

Abstract/ Executive Summary:

Water and salt soluble proteins of flag leaf in 100 and 120-day-old rice (BR6) plants were extracted and analyzed in 10 and 12.5 per cent SDS-PAGE. The detected number of peptide bands were 52 and 47 in 100 and 120-day-old leaves, respectively. The MW of the peptides varied from 12 to 200 kD. In 10-day-old plants the highest number of bands were mostly distributed in lower MW zones. In 120-day-old plants the highest number of bands were in middle MW zones. Out of all the major bands, 56 kD position peptide showed most abundant and drastically decrease in 120-day-old plants. Some pep tides also showed less intense during ageing. The other intense peptides were, 5.25,15.50,19.75,20.23,24, 28.50,29,41,42,66, 100 and 150 kD positions. Some peptides appeared in 100-day-old plants were not found in 120-day-old plants. At the same time some new peptides appeared in 120-day-old plants.

Objectives:

To find out at isolating and separating the water and salt soluble leaf proteins at the early and late reproductive phases of rice plants.

Methodology:

Rice (Oryza sativa var. indica L. cv BR6) was used in this study. Only the flag leaves at early and late stages were collected just above the leaf sheath at the age of 100-days (at the initial stage of flowering) and 120 days (senescence stage). Soluble protein was extracted according to the procedure. Sodium dodecyl sulfate (SDS) was avoided during extraction. The extracted protein was separated by 10 and 12.5 % SDS-PAGE (Sodium dodecyl sulfate polyacrylamide gel electrophoresis) according to the procedure. Protein solution containing about 200 μg/ml was mixed with equal volume of sample buffer and charged. A constant current of 220 v and 30 mA were used during the electrophoresis of gel. Coomassie brilliant blue was used for staining the gel. The destined (50 Me-OH : 10-GAA : 40 H₂O) gels were analyzed on incandescent light. The markers and the MW were urease-240 kd; bovine serum albumin (BSA) - 66 kD, chicken egg albumin (CEA)- 45 kD, carbonic anhydrase (CA)- 29 kD and a-Iactalbumin-14.2 kD.

Achievement/Findings:

It is indicated that leaf senescence involves not only the turning off some vital genes but also temporary and sequential inactivation of senescence related genes. For the identity of nuclear genes necessary for leaf senescence and the degradation of RUBlSCO enzyme study of the flag leaf of rice plant can be chosen as an experimental plant.
