Characterization of K+ -stimulated Adenosine Triphosphatase (k+ -atpase) Activity of Corn Roots

Abstract/ Executive Summary:

AT Pase activity of the roots of corn seedlings (Zea mays L. Bukovinskaya- 3 ) was stimulated by monovalent cations with the order of effectiveness being K+≥ Rb+>NH+Na+>Li+. Potassium- stimulated AT Pase activity was substrate specific, independent of Mg++ and showed no synergistic response to Na+. Maximum enzyme activity in presence of K+ was at pH 6.0 but higher K+- stimulation was at pH 3.5 and 5.0. K+- AT Pase activity of the corn roots was found associated mostly with the high speed cell membranes (18000- 127000 rpm pellets) sensitive to ouabain, N', N'- dicyclohexyl carbodiimid. kinetin and K+ concentration of the nutritional solution. The kinetic data of K+ - concentration were complex and followed the kinetics of negative cooperative.

Objectives:

To investigate the properties of K + - AT Pase of the roots corn seedlings.

Methodology:

Thoroughly washed corn seeds (Zea mays L. Bukovinskaya- B ) were soaked in hot water (60- 70 ? ) at room temperature for 2h and then allowed to germinate on moist fitier paper at 25°C for 2 days. The young seedlings then were transferred to Chesnokov's nutritional solution, pH 5.6- 5.8 and were grown in the dark at 25°C for 4 to 8 days old etiolated -or in the light for 10. to 20 days old green seedlings. The growth medium was aerated and changed after a regular interval of time. Root sections (20 mm, discarding 10 mm from the tip) from 4 days old seedlings or intact whole roots from 8 to 20 days were taken for enzyme preparation. Tris, bovine serum albumin. polya- crylamide and sodium salts of AMP, ADP, ATP, GTP, CTP, UTP, or Reanal, Hungary; Sodium salts of B- GP of Merck, FRG; sephadex g- 25 fine of Pharmacia, Sweden; kinetin and adenine of Chemapol, Czechoslo- vakia ouabain, dicyclohexilcarbodii- mid (DCCD) and chemically pure KCI, RbCl, NaCl, LiCI, NH₄CI; sucrose, EDTA- Na₂, amidol, ammonium molybdate and others of USSR; Dawex 50 (Zeo- karb 226) of Permudit company, UK and millipore ion exchange cartidge, CDHB 02202 of Bedford, USA Were used in the present work.
