Screening of Field Pea Genotypes Against Salinity Stress Under Laboratory Condition

Abstract/ Executive Summary:

Thirty field pea genotypes were tested against varying levels of salinity (0, 5, 10 and 15 dS m^-1) in Hoagland solution under laboratory condition at Plant Physiology Division, BARI during 2013-2014 to find out the salt tolerant genotypes at seedling stage. Distilled water (0 dS m^-1) was used as control. Germination percentage, seedling root and shoot length, root and shoot dry weight and vigor index were found affected by different levels of salinity. The genotypes BD-4159, BD-4150, BD-4163, BD-4161, BD-4152, BD-4160, BD-4167 and BD-4151 showed better performance up to 10.0 dS m^-1 in terms of germination percentage, seedling growth, and vigour index. These genotypes could be selected for further investigation under pot culture.

Objectives:

To find out the salt tolerant field pea genotypes at seedling stage

Methodology:

The experiment was conducted from December 24, 2013 to February 12, 2014 at Laboratory of Plant Physiology Division, Bangladesh Agricultural Research Institute (BARI). Thirty genotypes of field pea (BD-4135, BD-4136, BD-4137, BD-4141, BD-4142, BD-4143,BD-4144, BD-4145, BD-4147, BD-4149, BD-4151, BD-4152, BD-4153, BD-4159,BD-4160, BD-4161, BD-4163, BD-4165,BD-4167,BD-4171, BD-4177, BD-4181, BD-4182, BD-4183, BD-4185, BD-4186, BD-4187, BD-4190, BD-4195) collected from Plant Genetics Resource Center (PGRC), Bangladesh Agricultural Research Institute (BARI), Joydebpur, Gazipur were used in the study. The experiment evaluated the germination and seedling growth of field pea genotypes at different NaCl salinity levels. The NaCl concentrations used were 0.0 (control), 5.0, 10.0, and 15.0 dS m^-1. The salt solution was prepared with desired amount of NaCl in distilled water. Hoagland solution was used as nutrient media with the salt solution. The PH of Hoagland solution was maintained at 6.0. Plastic pots were used in the experiment with a diameter of 10 cm, height was 9 cm and arranged in a completely randomized design (CRD) with three replications. Each pot was contained 500 ml of the respective treatment solution. Twenty seeds were sown on the plastic pots having cotton gauge which covered the lid with small holes through which the roots can reach the solution. The germination count was taken after 72 hours of setting seeds. A seed was considered to be germinated when both the plumule and the radicle emerged > 0.5cm. After 15 days, the shoot and the root length of ten randomly selected seedlings from each replication were measured.

Achievement/Findings:

From this experiment it might be concluded that seedling growth of field pea genotype is decreased with the increase in salinity level and drastically decreased at 15 dS m^-1. The genotypes BD-4159, BD-4150, BD-4163, BD-4161, BD-4152, BD-4160, BD-4167 and BD-4151 showed better performance up to 10 dS m^-1 on the basis of germination (%), seedling growth and vigour index.
