The Effects of Casein and Amino Acid Diets on the Growth and Amino Acid Composition of Tilapia Fingerlings

Abstract/ Executive Summary:

A protein diet containing casein as the sole nitrogen source and three amino acid diets of similar composition were fed for four weeks to small Tilapia, Sarotherodon zillii. The effects of deitary casein and free amino acids on growth parameters, protein content, and free and bound amino acid compositions of the fish were studied. Higher growth rate was obtained with the fish on casein diet than with those on the amino acid diets. The lower growth rates with the amino acid diets were associated with an increase in the moisture and protein contents. The levels of free amino acids were reduced in the fish after feeding the amino acid diets. The levels of bound amino acids, however, were similar with both the casein and amino acid diets.

Objectives:

To determine and compare the concentrations of free and bound amino acids in tilapia, Sarotherodon zillii after feeding a purified protein diet and amino acid test .dlets of similar composition.

Methodology:

The complete amino acid diet was formulated to contain nearly the same amino acid com- position as the casein diet. All the diets were made iso- nitrogenous and isocaloric. A 11 the amino acids used were in the form, The vitamin and mineral mixtures were prepared . Amino acid diets deficient in cystine and tryptophan were prepared by deleting the respective amino acid from the mixture and replacing it with an equal amount of alanine. The diets were adjusted to pH 6.7±O.2 with 3 N NaOH. The diets were kept frozen at 10^oc until used. Experimental fish and feeding method: This study was conducted in Kagoshima University, Japan using tilapia fingerlings obtained from local hatchery. The fingerlings having a mean wt. of 1.62 g were acclimated for 2 weeks, the first week to a commercial diet and the second to the experimental diet at the standardized laboratory conditions. Twenty fingerlings were released into each aquarium of 30 X 30 X 60 em size, water was aerated and circulated at 400 mlrmin, through activated charcoal filter to remove imputes and reduce ammo- nia levels. The water temperature in the aquar i- was maintained at 25 ± 1°C. ({be fish were fed the experimental diets at the rate of 10% (on a wet weight basis ) of their total biomass per day in three equal feedings. The feeding experiment was continued for a period of four weeks.
