A Source of Heritable Nature of Colchicin Induced Variation in Inbred Lines of Wheat

Abstract/ Executive Summary:

Ten purely homozygous lines of young wheat seedlings were treated with 0.2% aqueous solution of colchicine for a period of two hours. The treated hexapioid plants were grown without further treatment. As a whole the experimental materiel showed two types of plant (1) colohicine treated hexapfoids end (2) control hexaploids. In an overall analyses the treatment induces changes in vegetative growth, flowering time and grain yield/ plant. Heritable nature of changes were observed In the self-seed generation following that in which the treatment was given. The changes was showed a directional bias which leads to an overall significant improvement in some important agronomic characters in wheat.

Objectives:

The aim of the present work was to test the hypothesis in 10 wheat varieties/advanced lines and genotypes in five important agronomic characters

Methodology:

It can be assumed that the materials were purely homozygous in nature. The materials are consisting of varieties/advanced lines/genotypes which are listed below : Aghrani. Kanchon, DSN-61, DSN- 72. MD-2329, Ad-66-1, Anza, SA-36, So-nalika and YC-17. The experiment was conducted at the Genetics and Plant Breedidg Farm. BAU, Mymensingh. In November 1990, wheat seeds after surface sterilization were placed on moist filter paper in petridishes. One week old wheat seedlings were treated with 0.2 percent aqueous colchicine for two hours at room temperature. The seedlings were treated by total immersion in colchicine solution and a second set of wheat seedlings were immersed in water as the untreated control. In November 1991 the seeds from the selfed plants were grown again without any further treatment. From the treated plants 30 plants from each of the 10 lines were grown on in 9- inch pots in the glasshouse together with same number of their control plantsThe harvested material was weighed to give fresh weight and was then dried in an oven at 80°c for 18 hours. The material was taken out from the oven and reweighed to give the dry weight, and the percentage dry matter was also calculated. Days to flower were also observed, which were taken as the date on which three spikelets were seen to have emerged from three separate inflorescence (expressed as number of days after 1 January). Straw/plant and grain yield per plant were determined after maturity. Plant belonging to the generation in which colchicine treatment was given are known as the (CT-0) generation and those derived from the selfed seed of the CT- 0 are known as the treatment generation-1 (CT-1 ).

Achievement/Findings:

The induced changes in most of t he characters are significant and transmitted through at least one self seed generation of sexual reproduction by selfing. The results drew attention due to the fact that the changes in the treated material made improvement in various important agronomic characters. Such changes may effectively be utilized in chalking out plant breeding programme for the improvement of this crop.
