Screening of Jute Mutants Under Different Levels of Salinity

Abstract/ Executive Summary:

A pot experiment was carried out at Bangladesh Institute of Nuclear Agriculture, Mymensingh during November 2002 to March 2003 to screen jute genotypes under salinity.The salinity treatments were 3 , 6 dSm^-1and a control where no salt was applied. The jute mutants CM-l07, P-26(5), CM-6l, CM-67, CM-452, BINA deshi pat-2, CM-443 and P-81(2) were used in this study. Salinity treatment influenced germination of the genotypes and two mutants P-81(2) and CM-67 and BINA deshi pat-2 were fully germinated under the salinity levels. Plant height, number of leaves, leaf area, root, stem, leaf weight, root-shoot ratio, total dry matter plant^-1, crop growth rate (CGR), chlorophyll-a, chlorophyll-b, total chlorophyll, Na⁺, K⁺ and Mg⁺⁺ contents in leaves were decreased with the increased in salinity levels compared to control. Relative growth rate, net assimilation rate, chlorophyll-a, chlorophyll-b, total sugar, reducing sugar and Ca++ content in leaves were unchanged with the salinity levels. Plant height, number of leaves, CGR, N₂ accumulation and total sugar in leaves were the highest in the jute mutant CM-443. Root dry weight, root shoot ratio and total dry matter were the highest in the mutant P-26(5). So, under salinity, CM-443 performed better followed by genotypes P-81(2) and CM-67.

Objectives:

To assess the effect of salinity levels on morphological and biochemical parameters of jute genotypes.
To find out salt tolerant genotype.

Methodology:

A pot culture experiment was carried out to screen jute genotypes on difilerent levels of salinity. The salinity treatments were 3 dSm^-1, 6 dSm^-1and a control where no salt was applied. The jute mutants CM-l07, P-26(5), CM-6l, CM-67, CM-452, BINA deshi pat-2, CM-443 and P-81(2) were used in this study. Salinity treatment influenced germination of the genotypes and two mutants P-81(2) and CM-67 and BINA deshi pat-2 were fully germinated under the salinity levels. Each pot contained 7 kg of soil collected from BINA farm. Urea, TSP and MP were applied at the rate of 60, 50, 60 and 20 kg ha^-1 respectively. Jute seeds were sown in pots on 13 April 2005. Amount of salt calculation for salinity development was done according to Michael (1978) and Ponnamperuma (1984). Salinity treatment was imposed at 20 days after sowing. Data on morpho-physiological attributes and yield were recorded. Data were analyzed statistically to find out the levels of significance using the MSTAT-C package program. Duncan’s Multiple Range Text (DMRT) was used to compare the differences of means.

CGR was also higher in the mutant CM-443. Chlorophyll-a and chlorophyll-b ratio and total N₂ accumulation in leaves were maximum in CM-443. Chlorophyll-a, chlorophyll-b were higher in CM-67 and total sugar was higher in CM-443. Under the salinity treatment CM-443 performed better than that of the other genotypes with respect to morphophysiological and biochemical parameters followed by genotypes P-81(2) and CM-67.
