Transferrin Types and Their Association with Milk Yield and Butter Fat Content in Dairy Cows

Abstract/ Executive Summary:

Ten transferring phenotypes consisting of four homozygotes. TfAA. TfBB, TFDD and TfEE and six heterozygotes, nAB HAD, TfAE, HBD. TfBE and TfDE have been detected in the blood sera of dairy cows. The animals were classified into ten groups according to the irtransferrin types and correlation was made between the transferring types and milk yield. lactation length and butter fat content of the animals The average per lactation milk yield of AA cows (1383.65 kg.), was found to be higher than other homozygOus types, TfBB (1205.06 kg.). TfDD (1066.43 kg.) and TfEE (1060.00 kg.) and the result was statistically insignificant. The increased milk yield was due to lager lactation length and not due to per day milk yield. No significant effect of transferrin types was observed on butter fat content. INTRODUCTION

Objectives:

To find out the transferrin genotypes and their association, if there be any, with milk yield and butterfat content in dairy cattle available in the Savar dairy farm.

Methodology:

Three hundred and twenty three dairy animals of the Central Dairy and Cattle Improvement Farm, Savar, Dhaka, were used in the investlqatton. The animals were physically healthy and managed under the same environmental conditions and nutritional status. Data on milk yield with butterfat content were recorded for four lactation periods of the cows. Blood sera obtained from the animals were preserved at-20°C in deep freeze until electrophoretic analysis was performed. Starch gel electrophoresis technique as described by Poulik (1957) was used with slight modification. The starch used in the system was extra ctad from pota . toes and hydrolysed in the laboratory by incubating per 100 gm of starch and 120011 of acetone with 8 011 of concentrated hvdroc'itoric acid at 40°C for one hour and15 minutes and washed with 1.51 of distilled.water in a Buchner funnel connected to an aspiration pump. This was then dried in an oven for about 72 hours at 40•C (Rahman et al. 1980). Gel was prepared from the hydrolysed lot by using tris buffer solution containing 9.2 gms of tris (hydroxy methyl) methyla- mine and 1.5 gm of citric acid in a litre of distilled w rter . The concentration of the starch gel was 13% w]», The gel thus prepared W3S poured into gel tray. The samples were inserted into the gels on pieces (3~x 6mm) of Whatman 2 filter paper No.3. Eight/nine samples were inserted into the gels and electro- phoresis was continued for 3 hours at an applied voltage of 350. Borate buffer containing 18.54 gm of boric acid and 2.8 9m5. of sodium hydroxide in a litre of distilled water was used in the electrode chambers.
