The morphogenesis and organ formation in tobacco pith was studied under different levals of kinetin and indole-acetic-acid (IAA). The epical explants yielded maximum callus at lower concentration of kinetin (.05 rnq/I) with a constant level of I AA (5 mg/I). With the increase of kinetin concentration. the total fresh weight of callus decreased. but bud and shoot formation initiated. The middle segments did not differentiate any bud and the callus formation was apparently very poor under all levels of kinetin with IAA. I n the bs sal segments. the callus ar.d bud formation were observed at maximum levels of kinetin and IAA.

The present lnvestiqaton was carried out with different levels of kinetin and at constant level of IAA on the morphogenesis and organ formation in tobacco pith callus cultured from apical, middle and basal seqments of stem

In order to prepare the explants about one meter tall tobacco stem. Nicotiana tobacum L. cv. 'Wisconsin 38', grown in a greenhouse, was cut at the base. After removal of leaves the ste n was cut into three pieces: apex, middle and base. Thereafter the stem segmen  were washed in water and disinfected by 70% ethanol and then kept for 15 minutes in 5% NaCIO to which 0.1 % Tween 20 was added. Cylinders of paren ihvrnatous pith ware then bored from apical, middle and basal segments by a sterile 5mm cork-borer. The extruded pith cylinders were then cut into 5mm pieces by discarding about 50101 from each of the end pieces as a precautions against any possible contamination. Murashige and Skoog's (1962, in- organic salts were supplemented with 0.4 mg/r thiamin-t+Cl , 100 mg/l meso inositol, 30 g, r sucrose, 8 gll agar and kinetin and IAA as showh in  1 I 2 131 4 I S"T6I7TB Kinetin(mg/I) lAd. Four pieces of apical, middle and basal pith seg- ments W.H6 then place.d in each flask. The cultures were then placed in a gro- wth-chamber under controlled environ- mental conditions where a constant 18- hour photoperiod with neutral light and a constant temperature of 21°C were maintained fora period of 721 weeks. The changes in cu Itures were observed at weekly intervals and the mean total tesh weiqt of the callus per flask was then measured at the end of 721 weeks. Medium No.8 was exclusively used for rooting of the developed shoots which were also placed under the same enviornamental condition  for a period of 2 weeks.
