Development of Rana Tigrina (daudin) in the Aquarium

Abstract/ Executive Summary:

Different metamorphic stages of Rana tigrina were studied under microscope. The time taken by various stages and the characters of tadpoles were noted. They took 42 to 52 days to become juvenile at room temperature. During 80 days of experiment with just hatched larvae, an average snoutvent length of 27.1±0.57 mm was obtained. Mortality was 34.83%.

Objectives:

To study the embryonic and larvae development of Rana tigrina in the aquarium.

Methodology:

Most of the mature frogs for present experiments were captured at night during the rainy seasons. Some of the frogs were captured when in clasping condition. They spawned in the aquarium. The eggs were equally divided into three aquaria containing about 740 eggs in each aquarium. The size of the aquarium was 44 x 30 x 22 cm, in which 10 cm water level was maintained. The tadpoles were thinned to 50 per aquarium and the water level was reduced to 3 cm and a portion of each aquarium was filled with sand as the tadpole gradually advanced the metamorphosis. The water was replaced every 48 hours. The embryonic development was studied from five samples (eggs) collected every thrity minutes after fertilization till the embryonic development was completed. The collected sample was put into the Frog's physiological solution (0.65% NaCI) for 2 to 3 minutes then these were studied under microscope. The desired ones were put in Bourn's fluid for 12 hours. Any cut damage or brittleness was considered undesirable. Then the samples were processed for slide preparation. The sections were cut at 10 μ by micro tome. The sections were taken on a glass slide with a drop of Mayer's albumen and the slides were put on the slide dryer having a temperature of about 50⁰C for stretching the sections, The sections were stained with haematoxylin and esion. For studying larval development five samples were collected every 24 hours for 52 days. The collected samples were preserved in 5% formalin. The slide prepared above and the desired samples from larvae were studied under microscope. Figures were drawn by camera lucida and the measurement were done by a graticuIe micrometer.

Achievement/Findings:

The duration of metamorphosis seemed to be dependent upon the temperature. Higher temperature enhances metamorphosis.
