The Reaction of Potato Cultivars to Black Leg/soft Rot Syndrome

Abstract/ Executive Summary:

Epidemiological aspects of the black leg/soft rot syndrome using conical potato eye plugs in controlled environment cabinet were studied. Temperature of 20⁰C is optimum for Infection by Erwinia carotovota subsp. atroseptica isolates and no infection occured at 5⁰C and 30⁰C. The rot developed in high and medium soil moisture levels was soft & massy, whilst that in the low moisture level was dry and tended to mummify. More soft rot was found to occur in young (0 day) than in old (12 days) eye plug sprouts, the reverse being the case in the development of black leg. Inoculation of eye plugs by tooth pick method proved to be useful in teasing the virulence for Erwinia subsp. tested. It was also useful in comparing the reaction of a number of cultivars In order of magnitude the cultivars Ulster Sceptre & Pentland Crown topped the list of susceptibility.

Objectives:

To elucidate the effect of temperature, moisture and sprout age of some potato cultivars and pathogen host relationship on soft rot/black leg syndrome.

Methodology:

The Erwirnia caratovora isolates which were used in this series of experiment. The potato tubers of different cultivate namely Ulster Seeptre. Pentland Crown, Pentland Squire, Pentland .Javaline. Desiree. Home Guard. Maris Piper. and Record were used to obtain eye plugs washed from soil. disinfected by dipping in 2.5% chloros (0 25% a. i. available chlorine) for 5 min. and air dried overnight. Tissue containing a single sprout was removed using a hemi-spherical scoop. Between each eye plug the scoop was disinfected by flaming in alcohol. The eye plugs were dipped in 1% Chloros for one min. and rinsed with distilled water. These were spread on moist filter paper and kept at a temperature of about 20⁰C for 3 days. During this the period cut surface of each eye plug become well suberised and they were then stored in cardboard box at 5°C until required for experimentation. A toothpick charged with an undiluted culture of Erwinie cerotovore was inserted 5 mm away from the eye plug sprout and to a depth of 5 mm. The end of the toothpick was snapped off bearing the portion carrying the inoculum inside. For finding the effect of temperature on blackleg/soft rot development. ten eye plugs in three replications of cv, Ulster Sceptre and cv. Pentland Crown were inoculated with the bacteria code no, 179/10 and planted in 2" x 2" jiffy pot containing sterile soil. The pots were kept on moist paper towels in a closed plastic box to provide high humidity, These were then kept at the incubation temperatures from 5 to 30⁰C, The diameter of rot was measured after 5 days of incubation. For effect of moisture on soft rot/ black leg development. ten eye plugs of cv. Ulster Sceptre replicated three times were inoculated by two E. stroseptlcs (code no. 179/10 and 761/14) and two E. carotovora subsp. carotovora (code no. 462/1 0 and 362/23) isolates. and incubated in three moisture regimes of 18,38 & 49% respectively. The inoculated eye plugs were planted in jiffy pots covered with potato compost soil and incubated at 20⁰C at 14/10 hours of alternating cycles of light and dark for 3 weeks, The light intensity was 17.000 lux. The soil moisture levels were maintained in the Jiffy pots as follows:- High-Water was placed in the tray so that the bottom of the pots wore submerged in the water (49%).
